Bulk RNA-seq analysis of primary human dermal fibroblasts from young and aged donors

ABSTRACT: Primary human dermal fibroblasts were obtained from four young donors (ages 23–33) and five aged donors (ages 64–72) through the San Diego Nathan Shock Center (SD-NSC) Human Cell Models of Aging Core. Cells were cultured under standard conditions and subjected to bulk RNA sequencing to investigate age-associated transcriptional changes. Libraries were prepared using polyA selection and sequenced on the Illumina NovaSeq 6000 platform. Differential gene expression analysis was performed using the HOMER pipeline.

Project description:The methylation profiles of bisulfite-modified DNA of human CD11b+ cells from healthy donors (HD) were compared with CD11b+ cells isolated from ovarian cancer patients (blood or ascitic fluid) using the Infinium HumanMethylation850 BeadChips (Illumina, Inc., San Diego, CA,). This platform allows the interrogation of >850,000 methylation sites per sample at single-nucleotide resolution, and comprises an average of 17 CpG sites per gene in the 99% of RefSeq genes. 96% of CpG islands are covered, with additional coverage in CpG island shores and the regions flanking them. The samples were hybridized in the array following the manufacturer’s instructions.

The miRNAs were quantified on custom Taqman Low Density miRNA Arrays from ThermoFisher Scientific; miRNA probes were consistent to those used in the UH2 discovery arrays.

Caring for Those Who Share: Improving the Health of Wisconsin Blood Donors

Project description:Blood donors are a precious community resource. Each month at Blood Center of Wisconsin (BCW), 200 men and 600 women over age 50 are turned away ("deferred") because of anemia. In those over 50, anemia may signal serious underlying disease such as colorectal cancer (CRC). CRC is the #3 cause of cancer death in Wisconsin with more than 1,000 deaths in 2006. Yet with proper testing CRC outcomes can be improved by early diagnosis. This project will focus on a population of deferred older blood donors to develop and test educational materials that will motivate donors to seek medical attention for their anemia, so that the underlying cause is diagnosed and treated. This project has the potential to lead to better CRC outcomes in Wisconsin blood donors.
