Off-Target RNA editing induced by RECODE in Mouse Quadratus Femoris Muscle

ABSTRACT: Programmable guide-RNA-dependent cytidine deaminase tools to convert cytidine-to-uridine (C-to-U) have limitations such as low editing efficiency, limited targetable sequence flexibility, and off-target RNA editing. Here, we report the editing activity of a novel guide-free C-to-U editing tool, named RECODE, based on RNA-binding pentatricopeptide repeat proteins. We show that RECODE, delivered via AAV, is functional in mice, demonstrating high editing efficiency and low off-target effects in quadratus femoris, a skeletal muscle. These results suggest its therapeutic potential for various diseases.
