Bulk RNA sequencing for gene expression profiling of iPSC-derived liver sinusoidal endothelial cells and arterial endothelial cells

ABSTRACT: Liver sinusoidal endothelial cells (LSECs) line liver sinusoids and regulate exchanges between blood and liver tissue, while maintaining hepatic vascular homeostasis through coagulation control, immune cell recruitment, and barrier function. To compare endotheial lineage markers, we performed bulk RNA-seq analysis of iPSC-derived LSECs (iLSECs) and arterial endotheial cells (iAECs). We here show that venous/LSEC programs in iLSECs (NR2F2, APLNR, CD36; STAB1, LYVE1, FCGR2B), whereas iAECs expressed higher pan-endothelial and arterial markers (PECAM1, CDH5; DLL4, HEY2, NRP1).
