Neuroprotective mechanisms of secreted factors from glial progenitor cells, obtained from human induced pluripotent stem cells

ABSTRACT: In this study, we investigated the mechanisms of neuroprotection of factors secreted by glial progenitror cells obtained from human induced pluripotent stem cells. We earlier got data on their neuroprotective action on the model of glutamate excitotoxicity in vitro, traumatic brain injury in vivo as well as proteomic analysis. The experiment was reproduced on cortical neuron cultures from Wistar rat pups (P0, 10 DIV). At 10 day of culturing, the drugs were added. The comparison groups were: extracellular vesicles, proteins of the 3-100 kDa fraction and total secretome. Total RNA was collected 4 hours after addiction of drugs for transcriptome analysis to identify signaling pathways activated in cells by drugs.
