VEGF-Induced Transcriptomic Alterations in A549 Cells

ABSTRACT: To investigate the mechanisms of cellular transformation, we performed transcriptome sequencing (RNA-seq) on VEGF-treated A549 non-small cell lung cancer (NSCLC) cells, with a focus on profiling changes in key target genes.

Global profiling of platinum drugs resistant genes in A549 cell lines

Project description:Non Small Cell Lung Cancer (NSCLC) causes the premature death of over 1 million people worldwide each year, but remains inadequately understood at the molecular level. To provide new insights for NSCLC treatment we performed a molecular characterisation of wild type and platinum drugs resistance in A549 cells. Transcriptome profiling revealed contrasting patterns of gene expression in sensitive and resistant cells and identified genes whose expression was highly correlated with the platinum drugs. Our results revealed a gene set of 15 transcripts whose expression was highly correlated with platinum-resistance in NSCLC A549 cell lines.

Transcriptomic analysis of two human non-small-cell lung cancer cell lines in response to SN1 methylating-agents treatment

Project description:Methylating agents of SN1 type constitute a widely used class of anticancer drugs, the effect of which on human non-small-cell lung cancer (NSCLC) has not been adequately studied. We thus studied the effect of N-methyl-N-nitrosourea (MNU), a model SN1 methylating agent, on two human NSCLC cell lines: A549 (p53wt) and H157 (p53null). We investigated the mechanism of MNU-induced cell death through a time course gene expression profiling study, 24, 48 and 72h following treatment. Two human NSCLC cell lines (A549, H157) were treated with MNU or DMSO for 24h, 48h, or 72h. 34 total samples were analyzed.

Knockdown of CUL3 and PTEN in A549 lung adenocarcinoma cells

Project description:Non-small cell lung cancers (NSCLCs) harbor thousands of passenger events that hide genetic drivers. Even highly recurrent events in NSCLC, such as mutations in PTEN, EGFR, KRAS, and ALK, are only detected in, at most, 30% of patients. Thus, many unidentified low-penetrant events are causing a significant portion of lung cancers. To detect low-penetrance drivers of NSCLC a forward genetic screen was performed in mice using the Sleeping Beauty (SB) DNA transposon as a random mutagen to generate lung tumors in a Pten deficient background. SB mutations coupled with Pten deficiency were sufficient to produce lung tumors in 29% of mice. Pten deficiency alone, without SB mutations, resulted in lung tumors in 11% of mice, while the rate in control mice was ~3%. In addition, thyroid cancer and other carcinomas as well as the presence of bronchiolar and alveolar epithelialization in mice deficient for Pten were also identified. Analysis of common transposon insertion sites identified 76 candidate cancer driver genes. These genes are frequently dysregulated in human lung cancers and implicate several signaling pathways. Cullin3 (Cul3), a member of an ubiquitin ligase complex that plays a role in the oxidative stress response pathway, was identified in the screen and evidence demonstrates that Cul3 functions as a tumor suppressor HumanHT-12 v4 Expression BeadChip Kit for human A549 lung adenocarcinoma cells with CUL3, PTEN, CUL3 and PTEN or no knockdown. A549 cells were stably transfected under Puromycin or Hygromycin selection to knockdown PTEN (SA Biosystems) or CUL3 (Open Biosystems) shRNAs. Genomic changes in low vs high metastatic A549 NSCLC cell line

Project description:Genomic changes in low and highly metastatic A549 cells were analyzed by 500K SNP arrays. A large number of genomic alterations were present in A549 cells but no significant differences were observed between the low or highly metastatic A549 cell lines. We generated a NSCLC line with highly increased propensity to form tumor nodules in murine lungs after intravenous injections. Extravasation and growth at a distant site are important parts of the metastatic process and we regarded these as a surrogate marker for in vivo aggressiveness and potential metastatic capability. A549 lung asdenocarcimona cell line with initially low metastatic potential was used for this purpose; these cells formed multiple small nodules in NOD/SCID mice after first i.v.-injection, round 1 (R1). Removal of tumor nodules from the lungs and subsequent re-injection led to a rapid increase in metastatic capacity. A highly aggressive phenotype which was stable over time was evident after three rounds (R3) of in vivo selection for the A549 cell line.

Here, we used RNA-seq for high-throughput profiling of transcriptomes in A549 cells transfected with AS-tDR-007333, in order to find differentially expressed genes regulated by AS-tDR-007333.
