Each sample was derived by pooling RNA from the relevant fat depot from 3 age-matched, male C57Bl6 mice.

T cells were isolated from tumors and draining inguinal lymph nodes of C57BL/6J male mice (7–8 weeks old) subjected to subcutaneous implantation of B16.gp33 melanoma cells, followed by adoptive cell transfer treatment with intravenous transfer of P14 and pmel T cells, and treatment with either isotype control antibodies or a combination of anti-PD-1 and anti-4-1BB antibodies. Samples were collected at day 14 post post-tumor implantation (1 day post-final antibody treatment). The study targeted all CD3+ T cell populations, including transgenic P14 and pmel T cells

Project description:This study is part of the immgenT Open Source Project, specifically from IGT33, which focuses on characterizing T cell populations in the uterus during Chlamydia trachomatis D and L2 infections. Mice received 1% transgenic NR23.4 (CD8) and NR1 (CD4) T cells intravenously 24 hours before infection to track antigen-specific CD8 and CD4 T cell responses. Mice were infected transcervically with C. trachomatis serovars D (UW-3/Cx) and L2 (434/Bu) (5x10E6 IFU) at day 0 and rechallenged at day 28 post-infection (5x10E6 IFU). T cells were isolated from the uterus of C57BL/6J mice at multiple time points: 5 and 15 days post-primary infection and 3 and 5 days post-secondary challenge. All animals were pre-treated with medroxyprogesterone acetate to synchronize estrous cycles.

Gene expression analysis of mouse lymphatic endothelial cells (LECs) from the subcapsular sinus (SS, afferent) and lymphatic sinus (LS, efferent)

Project description:Afferent lymphatic vessels bring antigens and diverse populations of leukocytes to draining lymph nodes, but efferent lymphatics allow only lymphocytes to leave the nodes. Despite fundamental importance of afferent vs. efferent lymphatics in immune response and cancer spread, molecular characteristics of these different arms of the lymphatic vasculature are largely unknown. In this work we report marked transcriptional differences between afferent and efferent lymphatic endothelial cells. Mouse inguinal, axillary, and brachial lymphnodes were collected and cryomolds prepared for laser-capture microdissection. Frozen sections were stained with various cell marker antibodies and micro-dissection performed to collect lymphatic endothelial cells. Total RNA was extracted from the cells and used in microarray analysis. Alternatively, popliteal, caudal, lumbar, inguinal, mesenteric, renal, axillary, brachial, deep and superficial cervical LN were enzymatically digested, the cells stained, and FACs sorted before RNA isolation.

From those mice, OT1 and OT2, not selected, positively selected, or negatively selected, were profiled.
