Whole-genome bisulfite sequencing (WGBS) for LKB1 overexpressing and NNMT knockdown A549 cells

ABSTRACT: Global hypomethylation is associated with senescence. Liver kinase B1 (LKB1) encoded by tumor suppressor gene serine/threonine kinase 11 (STK11) plays an important role in cellular senescence. LKB1 deficiency results in global DNA hypomethylation in lung adenocarcinoma accompanied by reduced DNA methyltransferase 1 (DNMT1) expression and elevated nicotinamide N-methyltransferase (NNMT) levels. However, whether LKB1 deficiency causes resistance to antitumor therapy by regulating cellular senescence through affecting global DNA methylation status remains unknown. Investigation by using A549 cells reveals that LKB1 deficiency inhibited DOX-induced senescence by upregulating NNMT to promote global DNA hypomethylation
