Functional division of insect blood cells in immunity by single-cell RNA sequencing

ABSTRACT: This study aimed to uncover the functional diversity and differentiation pathways of hemocytes in Spodoptera exigua by applying single-cell RNA sequencing (scRNA-seq). The objective was to classify hemocyte subtypes beyond traditional morphology and identify distinct gene expression profiles associated with immune responses, differentiation, and physiological functions.

Project description:The Spodoptera genus is defined as the pest-rich genus because it contains some of the most destructive lepidopteran crop pests, characterized by a wide host range. During feeding, the caterpillars release small amounts of oral secretion (OS) onto the wounded leaves. This secretion contains herbivore-induced molecular patterns (HAMPs) that activate the plant defense response, as well as effectors that may inhibit or diminish the plant’s anti-herbivory response. In this study, we explored the protein components of the OS of two Spodoptera species, Spodoptera exigua and Spodoptera littoralis.

A single-cell survey of Drosophila blood

Project description:Drosophila blood cells, called the hemocytes, are composed of three distinct cell populations: plasmatocytes (PM), crystal cells (CC), and lamellocytes (LM). These cell types have been extensively characterized based on the expression of a few marker genes and cell morphologies, which are inadequate to classify the complete hemocyte repertoire. Further, intermediate states along the course of maturation of these cell types have been poorly characterized. Here, we used single-cell RNA sequencing (scRNA-seq) to map the Drosophila larval hemocytes across different inflammatory conditions. We identified all known cell types, resolved PMs into different states based on the expression of genes involved in cell cycle, antimicrobial response, and metabolism together with the detection of CC and LM intermediates. Further, we discovered rare cells within CC and LM populations that express the fibroblast growth factor (FGF) ligand branchless (bnl) and receptor breathless (btl), respectively. We demonstrate that both bnl and btl are required for mediating effective immune responses against parasitoid wasp eggs in vivo, highlighting a novel role for FGF signaling in inter-hemocyte crosstalk. Our scRNA-seq analysis reveals the diversity of hemocyte populations and provides a rich resource of gene expression profiles for a systems-level understanding of their functions.

Solanum dulcamara's response to insect eggs comprises ovicidal hydrogen peroxide production

Project description:Plants can respond to insect oviposition but little is known about which responses directly target the insect eggs and how. Here, we reveal a mechanism by which the bittersweet nightshade, Solanum dulcamara, kills the eggs of a generalist noctuid herbivore. The plant responded at the site of oviposition by Spodoptera exigua with neoplasm and chlorotic tissue formation, accumulation of reactive oxygen species and induction of defence genes and proteins. Transcriptome analysis revealed that these responses were reflected in the transcriptional reprogramming of the egg-laden leaf. The plant-mediated egg mortality on S. dulcamara was not present on a genotype lacking chlorotic leaf tissue at the oviposition sites on which the eggs are exposed to less hydrogen peroxide. As exposure to hydrogen peroxide increased egg mortality, while catalase supplementation prevented the plants from killing the eggs, our results suggest that ROS formation directly acts as an ovicidal plant response of S. dulcamara.

Mitochondrial perturbations in immune cells enhance cell-mediated innate immunity in Drosophila

Project description:Mitochondria participate in various cellular processes including energy metabolism, apoptosis, autophagy, ROS production, stress responses, inflammation and immunity. We investigated the effects of immune tissue specific mitochondrial perturbations on immune responses at the organismal level. Drosophila melanogaster model was utilized to knockdown genes from oxidative phosphorylation (OXPHOS) complexes cI-V, by targeting the two main immune tissues of Drosophila, the fat body and the immune cells (hemocytes). While fat body targeted OXPHOS perturbations caused detrimental effects on the viability and immunity, hemocyte specific perturbations led to enhanced immunocompetence of the host. This was accompanied by the formation of melanized hemocyte aggregates (melanotic nodules), a sign of activated cell-mediated innate immunity. Furthermore, hemocyte specific OXPHOS perturbations caused proliferation and immune activation of the hemocytes, resulting to a similar hemocyte profile as seen upon infection, and the animals ultimately had an enhanced immune response when infected with a parasitoid. Our data shows that hemocyte targeted OXPHOS perturbations cause mitochondrial membrane depolarization and upregulation of genes associated with mitochondrial unfolded protein response, including aerobic glycolysis and reactive oxygen species. Overall, we show that while the effects of mitochondrial perturbations on immune responses are highly tissue specific, mild mitochondrial dysfunction can be beneficial in immune-challenged individuals and is causing variation in infection outcomes between individuals.
