Single-cell sequencing technology reveals the characteristics of the tumor microenvironment in gastric cancer and its peritoneal metastases

ABSTRACT: To analyze the tumor microenvironment characteristics of gastric cancer and its peritoneal metastases, we collected fresh surgical specimens pathologically confirmed as gastric cancer and peritoneal metastases, and performed single-cell sequencing.

Project description:Peritoneal carcinomatosis is a frequent finding in patients with primary gastric cancer, and it is associated with a poor prognosis. A major mechanism in peritoneal carcinomatosis is the dissemination of cancer cells into the abdominal cavity, mainly in diffuse gastric adenocarcinoma. The features that enable diffuse primary gastric tumours to develop peritoneal dissemination have been little investigated and are only incompletely understood. We therefore compared the gene expression profile in patients with diffuse primary gastric cancer with and without peritoneal carcinomatosis. Specimens from consecutive gastric cancer patients with and without peritoneal carcinomatosis were investigated using oligonucleotide microarrays. Keywords: Disease state analysis

Proteomics characterization of human peritoneal tissue

Project description:To identify the proteome of human peritoneal tissue, 43 FFPE and 5 fresh frozen peritoneal tissue samples at healthy or non-cancerous state were collected and subjected to mass spectrometry analysis. The resulting findings represnt a baseline catalog of the proteomic composition of the peritoneum, as a comparison dataset for future studies focused on alterations in pathologic states such as peritoneal carcinomatosis.

Non-invasive liquid biopsy based on transcriptomic profiling for early diagnosis of occult peritoneal metastases in locally advanced gastric cancer

Project description:Gastric cancer (GC) remains one of the leading causes of cancer-related death worldwide, especially in East Asian countries. Despite advances in treatment, patients with locally advanced gastric cancer (LAGC) still have a poor prognosis due to the high incidence of metastasis and recurrence. Peritoneal metastasis is particularly occult and difficult to detect by conventional imaging techniques at an early stage. Existing imaging methods lack high sensitivity and are difficult to detect early metastatic lesions. Although diagnostic laparoscopy has high sensitivity, it is invasive and difficult to use routinely. Therefore, innovative diagnostic methods are urgently needed. Transcriptomics methods can provide a comprehensive understanding of the biological state of cancer, especially showing potential in the early detection of GC and peritoneal metastasis. Our objective is to provide a more sensitive diagnostic method in the latent stage of peritoneal metastasis by identifying the RNA expression pattern of cancer cells.

Identification of Novel Paracrine Therapeutic Targets in Peritoneal Metastases through Integrative Proteotranscriptomic Analysis of Ascites

Project description:The presence of ascites in peritoneal metastases (PM) portends a poor prognosis for all intra-abdominal malignancies. However, the molecular ligands within ascites and their biological relevance remain unclear. We report a comprehensive proteomics analysis of cell-free ascites in PM, including 160 malignant PM ascites covering a spectrum of 17 histological origins and 15 ascites of benign origin. This is the first large-scale proteomic analysis of cell-free ascites across diverse histological subsets of PM that can be harnessed for discovery of novel therapeutics and understanding of PM biology.

The single cell transcriptional landscape of human lung adenocarcinoma (primary tumours and metastases)

Project description:By transcriptionally profiling single cells from patients spanning different stages of lung adenocarcinoma progression, we provide evidence of epithelial regeneration in untreated primary tumours and a continuum of phenotypic states spanning key stages of embryonic development and lung morphogenesis in metastases. We transcriptionally profiled 41,384 single cells obtained from fresh surgical human samples taken from non-tumour-involved lung (n = 4), primary lung adenocarcinomas (n = 8; 7 untreated and 1 post neo-adjuvant chemotherapy), as well as brain (n = 3), bone (n = 1), and adrenal (n = 1) lung adenocarcinoma metastases. These samples were derived from patients spanning various stages of tumour progression without enrichment for a specific cell type, such that the entire tumour and its microenvironment were sampled in an unbiased manner. Data from all patients were merged to create a global cell atlas of the normal lung, primary tumours, and metastases.

Gene expression profile prospectively predicts peritoneal relapse after curative surgery of gastric cancer

Project description:Purpose: Our study aimed to disclose the specific gene expression profile representing peritoneal relapses inherent in primary gastric cancers and to identify patients at high risk of peritoneal relapse in a prospective study on the basis of the molecular prediction. Experimental Design: RNA samples from 141 primary gastric cancer tissues after curative surgery were profiled using oligonucleotide microarrays covering 30,000 human probes. Firstly we constructed molecular prediction system and validated the robustness and prognostic validity of the analysis by 500 times multiple random sampling in 56 retrospective set consisting of 38 relapse free and 18 peritoneal relapse patients. Secondly we applied this prediction to 85 prospective set to assess the predictive accuracy and prognostic validity. Results: In retrospective phase, 500 times multiple random sampling analysis yielded 68% predictive accuracy in average and 22 gene expression profile associated with peritoneal relapse was identified. This prediction could identify significantly poor prognostic patients. In prospective phase, the molecular prediction yielded 76.9% overall accuracy. Kaplan–Meier analysis with peritoneal relapse free survival showed a significant difference between ‘good signature group’ and ‘poor signature group’ (Log-rank p=0.0017). Multivariate analysis by Cox regression hazards model revealed that the molecular prediction was the only independent peritoneal relapse prognostic factor. Conclusions: Gene expression profile inherent in primary gastric cancer tissues can be useful to predict peritoneal relapse prospectively after curative surgery and individualize postoperative management to improve the prognosis of advanced gastric cancers.
