PFOA impairs host immune response in viral infection

ABSTRACT: We performed scRNAseq on spleen cells pooled from 4 mice per group using the 10x Genomics scRNA-seq Flex assay, following the protocol outlined by the Chromium Fixed RNA Profiling Reagent Kit. Single cell sequencing of splenic lymphocyte revealed that PFOA. Furthermore, we leverage the lymphocytic choriomeningitis virus (LCMV) mouse model to quantify virus-specific immunity after PFAS exposure. PFOA exposure impaired the viral clearance and viral specific T cell response. Single cell RNA-seq analysis revealed downregulation of TNF-a signaling via NF-kB and interferon gamma response in effector CD8 and CCL5hi CD8 T cells. exposure modulates the immune signatures in B and T cells, with decreased anti-viral genes and increased autoimmune related genes.
