Project description:Background: We utilized a CRISPR-Cas9 screening library to examine the functions of tumor-related genes. Among these, we identified the transcription factor ZBTB7B, which overexpressed in breast cancer cells and significantly inhibited breast cancer growth in the brains of nude mice. Based on this finding, we hypothesized that ZBTB7B might exert similar effects on glioma, a possibility not yet explored in the literature. This study aims to evaluate the effects and mechanisms of ZBTB7B in glioma progression. Methods:The U118 and the GL261 cell line , both overexpressing ZBTB7B, were generated via lentiviral transduction. The impact of ZBTB7B on glioma growth was assessed using in vitro cell culture and in vivo intracranial inoculation. Downstream targets regulated by ZBTB7B were identified through transcriptome sequencing, PCR arrays, ELLA assays, and Western blotting. Cell apoptosis and the cell cycle were analyzed via flow cytometry. Multicolor immunofluorescence staining was performed to evaluate the expression and localization of ZBTB7B in human and mouse glioma tissues. Findings:Overexpression of ZBTB7B significantly inhibited glioma cell survival both in vitro and in vivo. Mechanistically, ZBTB7B upregulated the expression of GPR17, which suppressed PKA phosphorylation, promoted the generation of reactive oxygen species, and ultimately induced apoptosis. Additionally, ZBTB7B regulated the chemokine CXCL10, facilitating the recruitment of CD4+ and CD8+ T cells to the tumor periphery. Human glioma tissue microarray analysis revealed ZBTB7B expression was significantly reduced or absent in grade II to IV glioma samples. Interpretation:ZBTB7B exhibits significant tumor-suppressive effects and holds promise as a potential molecular target for glioma therapy.

The control (naive) animals were left untreated. The brain hemispheres from the control animals, or the tumor-bearing hemispheres were dissected, gently dissociated into cells and the microglia (CD11b+CD45low cells) were sorted out.

Project description:Glioblastoma (GBM) is a lethal brain tumor without effective treatment options. Data about proteomics in glioma is sparse and it has not yet been integrated into the routine diagnostic work-up or exploited to find potential actionable targets to prevent GBM-induced immune evasion mechanisms and recurrence. We performed proteomic analysis of diffuse glioma samples classified into six subgroups defined by DNA methylation and patient-paired primary and recurrent samples in order to i) provide an inventory of proteins commonly or individually overexpressed in each of the four HGG subgroups compared to the LGG and deduce relevant pathways for each subgroup and ii) identify specific proteins involved in tumor recurrence after surgical removal.
The first study revealed largest differences between low-grade (LGG) and high-grade gliomas (HGG), with major changes observed in proteins involved in active calcium-dependent vesicle trafficking (S100 proteins and annexins), effectors of the immunological synapse, proteins involved in Retinoid acid signalling, and proteins involved in extracellular matrix, cytoskeleton, and cell adhesion. Comparative proteomic analysis between LGG and the HGG subgroups also showed consistency with glioma grading.
The second study identified FcGamma receptors on activated microglia and complement components as major players inducing short relapsing GBM tumors.
In conclusion, specific pathways and proteins represent potential targets to control progression of the distinct subgroups.

Project description:The discovery of the oncometabolite 2-hydroxyglutarate in isocitrate dehydrogenase 1-mutated (IDH1-mutated) tumor entities affirmed the role of metabolism in cancer. However, large databases with tissue metabolites that are modulated by IDH1 mutation remain an area of development. Here, we present an unprecedented and valuable resource for tissue metabolites in diffuse glioma and their modulations by IDH1 mutation, histology, and tumor treatments in 101 tissue samples from 73 diffuse glioma patients (24 astrocytoma, 17 oligodendroglioma, 32 glioblastoma), investigated by NMR-based metabolomics and supported by RNA-Seq. We discovered comparison-specific metabolites and pathways modulated by IDH1 (IDH1 mutation status cohort) and tumor entity. The Longitudinal investigation cohort provides metabolic profiles of untreated and corresponding treated glioma samples at first progression. Most interestingly, univariate and multivariate cox regressions and Kaplan-Meier analyses revealed that tissue metabolites correlate with progression-free and overall survival. Thus, this study introduces potentially novel candidate prognostic and surrogate metabolite biomarkers for future prospective clinical studies, aiming at further refining patient stratification in diffuse glioma. Furthermore, our data will facilitate the generation of so-far-unanticipated hypotheses for experimental studies to advance our molecular understanding of glioma biology.
