The gene expression of human adipose-derived stem cells treated with butyrate

ABSTRACT: Adipose-derived stem cell (ASC)-conditioned medium shows therapeutic promise for cardiac repair via paracrine mechanisms. Moreover, in our previous report, cellular priming with butyrate (BA) is a promising approach for modulating the secretory phenotype of ASCs. Therefore, the comprehensive gene expression analysis was conducted on human ASC preconditioned with butyrate.

Transcription profiling of human H1 hESC exposed to butyrate

Project description:To assess the effect of sodium butyrate exposure on human ESC grown without culture support for self-renewal (I.e. without conditioned medium and added bFGF) - three groups were compared - H1 culture in feeder conditioned medium vs without conditioned medium in 0.2 mM sodium butyrate vs. grown in sodium butyrate for 4 passages followed by return to conditioned medium conditions for 3 passages. The three groups were grown in triplicate and compared on Agilent whole human genome array Experiment Overall Design: H1 hESC were grown under 3 conditions: A. 48 passages total, the last 9 passages off of feeders on Matrigel in feeder conditioned medium B. 48 passages total, within the last 9 passges, #39-41 were in conditioned medium and #42-48 were without conditioned medium and added bFGF but with 0.2 mM sodium butyrate on Matrigel C. 49 passages total, within the last 10 passages #39-41 were in conditioned medium, #42-46 in 0.2 mM sodium butyrate (without feeder of bFGF support) and #47-49 back to conditioned medium + bFGF. The RNA from these cells were compared via Agilent whole human genome array.

Proteome analysis of adipose-derived stem cells cultured under clinically relevant conditions in a wound healing perspective

Project description:There is a large body of evidence supporting the beneficial role of adipose-derived stem cells (ASCs) in tissue repair and regeneration. These effects appear to be mainly mediated by paracrine signaling pathways and enhanced during hypoxia. Mass spectrometry (MS) represents a valuable tool for proteomic profiling of cultured ASCs, which may help revealing the identity of the factors secreted by the cells under different conditions. However, serum starvation essentially required to obtain samples compatible with secretome analysis by MS can have significant influence on ASCs. Here, we present a novel and optimized culturing approach based on the use of a clinically relevant serum-free formulation, which was used to assess the effect of hypoxia on the ASC proteomic profile.

Influence of VEGF stimulation and serum deprivation on gene activation patterns of human adipose tissue-derived stromal cells

Project description:ASCs cultured in complete medium, ASCs cultured in serum serum-deprived medium, and ASCs stimulated with VEGF in serum-deprived medium were compared. Using microarray analysis, gene expression from the whole genome was compared between conditions. Compared to ASCs in complete medium, expression of 190 and 108 ASC genes were significantly regulated altered by serum deprivation and serum deprivation combined with VEGF, respectively. No significant differences in gene expression patterns between serum-deprived ASCs and serum-deprived ASC combined with VEGF stimulation were found. Genes most prominently and significantly up-regulated by both conditions were growth factors (IGF1, BMP6, PDGFD, FGF9), adhesion molecule CLSTN2, extracellular matrix related proteins like matricellular proteins SMOC2, SPON1 and ADAMTS12, and inhibitors of proliferation (JAG1). The most significantly down-regulated genes included matrix metalloproteinases (MMP3, MMP1), and proliferation markers (CDKN3) and GREM2, - a BMP6 antagonist.
