Identification of Cox5b and Sdhb as Diagnostic Biomarkers for Obesity via Calorie Restriction Modeling and Immune Cell Infiltration Analysis

ABSTRACT: This study aimed to identify key genes linked to obesity using a calorie-restricted mouse model and to explore the role of immune cell infiltration in disease pathology. Subcutaneous adipose tissue from mice underwent transcriptome sequencing, revealing 6,197 differentially expressed genes (DEGs; 2,837 upregulated, 3,360 downregulated) between the experimental (Group D) and control groups (Group N). Hub genes Cox5b and Sdhb were identified through intersection of WGCNA and PPI network analyses. Immune cell infiltration profiles differed significantly between groups, with Cox5b/Sdhb expression correlating strongly with specific immune populations. Validation in human PBMCs confirmed downregulation of both genes in obesity (p<0.01). ROC curves demonstrated high diagnostic accuracy for Cox5b (AUC=0.85) and Sdhb (AUC=0.93), suggesting their utility as obesity biomarkers. These results reveal novel regulatory mechanisms involving mitochondrial genes and immune dynamics in obesity.
