CD4+ T-Cells Mediate CART Immune-Related Adverse Events (CirAE) Following BCMA CART Therapy

ABSTRACT: We employed single-cell RNA sequencing (scRNA-seq) using the 10x Genomics platform to study the immune landscape of CAR T cells in the peripheral blood (PBMC) and cerebrospinal fluid (CSF) of patients treated with Cilta-cel who developed CAR T-cell immune-related adverse events (CiRAE). CAR T cells were captured and profiled across both compartments. This study offers insights into how CAR T cell phenotypes and functional states diverge between the periphery and central nervous system during inflammatory neurotoxicity.
