Adenosine - a regulator of human testicular peritubular cells


ABSTRACT: Extracellular purines, particularly ATP and adenosine, are key regulators of physiological and pathophysiological processes throughout the human body. Despite their systemic importance, the specific roles of purinergic signaling in the human testis and their potential relevance for male fertility, are not well studied. As these processes cannot be readily studied in vivo, human testicular peritubular cells (HTPCs), which form the wall of seminiferous tubules, provide a unique and accessible in vitro model. HTPCs were isolated from testicular samples and cultured. Transcriptional profiles of adenosine-treated cells were analyzed using scRNA-seq, revealing strong upregulation of IL33, STAR, HIF1A, and others. Pathways linked to cell communication, migration and inflammation were enriched in HTPCs following adenosine treatment.
