MBD3 regulates thymic Treg precursor cell development by modulating CD25 expression [RNA-seq]

ABSTRACT: RNA-seq was performed on thymocytes, including double-positive (DP), immature CD4 single-positive (ImCD4SP), pre-tTreg (precursor thymic Treg cells), and mature tTreg cells, from wild-type (WT) and Mbd3 conditional knockout (CKO) mice, in which MBD3 exon 2 is specifically deleted in T cells via Cre recombinase. The data capture gene expression changes associated with Treg lineage specification in the thymus, revealing transcriptional dysregulation upon MBD3 loss, particularly affecting later stages of tTreg development, including cell proliferation, apoptosis, response to stimulation, and expression of Treg-associated genes. These RNA-seq datasets provide insight into how the common factor MBD3 selectively regulates thymic Treg development at the transcriptional level.
