Project description:Cervical cancer (CC) is a malignancy characterized by persistent HPV infection and tumor microenvironment remodeling. To explore the molecular characteristics of plasma extracellular vesicles (EVs) in CC, we performed miRNA sequencing on plasma EVs from 3 CC patients and 3 healthy controls. Differentially expressed miRNAs were identified, revealing potential biomarkers and providing insights into early diagnosis and therapeutic intervention.

Project description:Cervical cancer (CC) remains a significant public health issue in low- and middle-income countries (LMICs), especially in Western sub-Saharan Africa and Nigeria. While global CC incidence and mortality have declined, these regions continue to face high rates due to inadequate screening and the high prevalence of HIV, which increases CC risk by promoting persistent HPV infections. This study aimed to identify DNA methylation (DNAm) biomarkers for cervical intraepithelial neoplasia (CIN) and CC in HIV-positive Nigerian women and to assess their potential for clinical risk prediction. From 2018 to 2020, 538 participants were recruited from Nigerian tertiary hospitals. Cervical tissue samples were analyzed for DNAm using the Infinium MethylationEPIC BeadChip array, and HPV genotyping was conducted via next-generation sequencing. An epigenome-wide association study revealed 24 significant DNAm biomarkers associated with CIN and CC. These biomarkers showed hypermethylation in tumor suppressor genes (e.g., PRMD8), hypomethylation in oncogenes (e.g., MIR520H), and aberrant methylation in genes related to HIV/HPV infection and oncogenesis (e.g., GNB5, LMO4, FOXK2, NMT1). A machine learning-based DNAm classifier achieved 92.9% sensitivity and 88.6% specificity in predicting CC risk, with higher risk observed in adjacent normal cervical samples from CIN/CC patients and HIV/HPV co-infected women. DNAm biomarkers offer a promising approach to enhancing CC screening and early detection, particularly for HIV-positive women in LMICs. The DNAm-based model developed in this study shows potential for more accurate CC risk stratification, highlighting the need for further optimization, validation, and implementation in low-resource settings.

Project description:<p>This study aimed to highlight the relationships between vaginal microbiome composition, metabolic alterations, and gene expression changes during cervical cancer. Multi-omics analysis for the healthy (Z group), abortion (AB group), HPV infection (HP group), and cervical cancer group (CC group), qRT-PCR and cytotoxicity assays were used for validation. The CC group had the highest vaginal community diversity with decreased Lactobacillus and increase in anaerobic bacteria. In the HP group, Bifidobacteriales are obvious markers. Metabolomic analysis revealed changes in lipid metabolism pathways, the metabolic pathways of the AB and HP group were similar. Lactic acid and α-linolenic acid were negatively correlated with Anaerococcus and Porphyromonas. There were 8 up-regulated genes at HPV infection. Key upregulated genes in the CC group, including CCL2, CDKN1A, and IRF1, showed negative correlations with α-linolenic acid levels. The distinct microbial and metabolite patterns could facilitate in the diagnosis and treatment of cervical cancer. </p>

Project description:Although Human papillomavirus infection is the main causal factor for cervical cancer (CC), there is data suggesting genetic factors could modulate the risk and progression of CC. Sibling studies suggest that maternally inherited factors could be involved in CC. To assess whether mitochondrial DNA (mtDNA) polymorphisms are associated to cervical cancer, HPV infection and HPV types, a case-control study was performed in the Mexican mestizo population. The polymorphism of mtDNA D-Loop was investigated in 187 cervical cancer patients and 270 healthy controls. D-loop was amplified from a blood DNA sample and analyzed by sequencing. HPV was detected and typed in cervical scrapes from both groups. mtDNA polymorphisms were compared in the whole samples and stratified by HPV types. The expression of 29 mitochondrial genes was analyzed in a subset of 45 tumor biopsies using the expression microarray ST1.0. The Amerindian haplogroup B2 increased the risk for CC (OR=1.6, 95% CI: 1.05-2.58) and showed an additive effect of 36% over the risk conferred by the HPV (OR=153, 95% CI: 65.4-357.5). The frequency of HPV 16, 18, 31 and 45 in cancer samples was similar in all haplogroups but one (D1). It showed a very low frequency of HPV16, any HPV18 and high frequency of HPVs 31, 45 and other types. Two mtDNA genes (MT-TD, MTTK) could be involved in the increased risk conferred by the haplogroup B2, since they were up-regulated exclusively in B2 tumors (p<0.05, t-test). These findings will contribute to clarify the importance of genetic factors in CC.

Project description:Majority of the patients with cervical cancer present with locally advanced cervical cancer (LACC), which is primarily treated using concurrent chemoradiation (CCRT) and lacks reliable predictive biomarkers. Extracellular vesicles (EVs) could potentially define the dynamic biological response to CCRT. However, the relationship between EVs and the therapeutic response to LACC is unestablished. This study aimed to determine the relationship of plasma EVs pre- and post-CCRT in 62 patients with LACC. EVs were isolated using ultracentrifugation (UC) with size exclusion chromatography or UC alone for proteomic analyses. The plasma particle concentration significantly increased post-treatment in non-responders. The viral control process was enhanced at 1 month and adaptive immune response increased at 3 months, following the standard clinical cut-off. Sparse partial least squares-discriminant analysis revealed 8, 13, and 19 proteins at diagnosis, 1 month, and 3 months, respectively, influencing the CCRT response. Among these, FIBG, TFR1, HBA, and FINC, are poor prognostic markers in the cancer genome atlas tissue gene expression database. The discriminant model demonstrated excellent specificity and negative predictive value. In conclusion, the validation of our model underscores its reliability in determining the responsiveness to CCRT, highlighting the potential clinical applicability of EVs in improving outcomes in LACC.

Project description:Here we performed single-cell RNA sequencing of more than 30,000 of cells from HPV16+ and HPV- cervical cancer patients. We comprehensively revealed the heterogeneity of both the malignant cell populations and immune lineages in HPV+ and HPV- CC, as well as the cellular crosstalks with potential relevance to tumor progression. Moreover, our results demonstrated that distinct interaction patterns in HPV+ and HPV- groups could mediate similar functions or pathways, and we further constructed a three-cell interaction resource.

Project description:Comparative RNA profiling between tumor cells and their secreted extracellular vesicles. Results revealed enrichment in genes involved in cellular migration and metastasis in extracellular vesicles, in agreement with their role as mediators of tumor progression. Mice were orthotoplically transplanted with MDA-MB-231 Breast Adenocarcinoma cells. Cells and extracellular vesicles (EVs) from the resulting tumors were isolated. EVs were characterized by electron microscopy and Nanoparticle Tracking Analysis before total RNA isolation for comparative analysis with cellular RNA. Three biological replicates were analyzed in (technical) duplicate.

Project description:Recurrent karyotypic abnormalities are a characteristic feature of cervical cancer (CC) cells, which may result in deregulated expression of important genes that contribute to tumor initiation and progression. To examine the role of genomic copy number alterations, we surveyed genetic lesions in CC utilizing single nucleotide polymorphism (SNP) array. We identified specific genetic alterations associated with CC. These data will be useful in identification of target altered genes, novel markers for predicting high risk precancerous lesions to invasive cancer, comparison of copy number alterations with gene expression changes can provide gene targets for pharmacologic intervention. We demonstrate specific regions of gene amplification (e.g., 11q22), copy number gains (e.g., 3q, 5p, and 20q), and deletions (e.g., 2q, 11q23) in the present study, which forma a framework for identification of critical genes in CC tumorigenesis. Project description:Background: Cervical cancer (CC) is a global health challenge, ranking fourth among cancers in women. Microbiome-metabolome interactions influence human papillomavirus (HPV) associated carcinogenesis, but specific microbial metabolites driving malignant progression remain undefined. This study aimed to identify potential biomarkers for distinguishing CC, and further explore their role in the progression of CC. Methods: Non-targeted metabolomics was employed to profile alterations in the vaginal microenvironment across clinical cohorts, including individuals with CC, individuals with cervical intraepithelial neoplasia (CIN), HPV-positive individuals, and HPV-negative individuals. Targeted metabolomics was then used to confirm the expression of 4-ethylbenzoic acid (4-EA) levels and its role in CC was explored using cell counting kit-8, 5-ethynyl-2'-deoxyuridine, colony formation, transwell, and wound healing assays. Proteomics was used to investigate the effects of 4-EA on CC cells. Results: The metabolic profiles of vaginal secretions in the CC group differed significantly from those in the other three groups. Untargeted metabolomics identified 27 CC-specific metabolites (VIP>2, P<0.05), revealing a marked elevation of 4-EA and its close relationship with vaginal microorganisms. Clinico-pathological correlations revealed progressive 4-EA accumulation across the cervical carcinogenesis stages. Additionally, 4-EA promoted the proliferation, migration, and invasion of CC cells in vitro. Proteomic reprogramming of CC cells following 4-EA treatment identified 14 highly expressed proteins associated with poor prognosis. Conclusions: This multi-omics investigation identified 4-EA as a novel candidate metabolite and a potential biomarker of CC. Identification of key proteins may provide new insights for interventions targeting the development of CC.
