ABSTRACT: CD34+ cell–derived megakaryocytes from four independent donors following treatment with PBS control, IFNα (1000 U/mL), or IFNγ (100 ng/mL). These data define donor-resolved interferon-responsive gene programs in human megakaryocytes under type I versus type II interferon stimulation.

Platelet Activation and a Platelet Biosignature Associates with Enhanced Cardiovascular Risk in Patients with Controlled Psoriasis.

Project description:The underlying mechanisms of atherosclerosis and strategies identifying high cardiovascular (CV) risk in psoriasis are incompletely understood. Platelet activity is increased in psoriasis and induces vascular dysfunction. We investigated the platelet phenotype and platelet transcriptome as one potential mechanism to explain CV risk in psoriasis. Psoriasis and controls underwent platelet aggregation and activation studies and platelet RNA sequencing to generate a psoriasis platelet transcriptomic score. The relationship between the platelet transcriptomic score and CV risk was assessed by arterial stiffness, coronary calcium, and longitudinally in an independent cohort of high CV-risk individuals undergoing lower extremity arterial revascularization (LER).

RNAseq of Huh7 and Huh7.5 cells treated with interferon alpha, beta or lambda.

Project description:The transcriptome of Huh7 and Huh7.5 was sequenced from cells treated with interferon alpha (IFNα), beta (IFNβ) or lambda (IFNλ) for 8 hours. Each treatment was compared to respective mock-treated cells in 6 pairwise comparisons, for a total of 43 samples classified in 12 experimental conditions. Cells were treated for 8h with 10ng/ml of Interferon-α2A, 5000 U/ml of Interferon-β1A or 100ng/ml of interferon-λ3 prior RNA extraction and library preparation. Mock-treated cells were used as control for each treatment. 3 to 4 biological replicates were analysed for each biological condition. Libraries for Illumina sequencing were constructed by polyA selection with the Illumina Kit. A paired end (2x100) run was performed.

Mechanical confinement prevents ectopic platelet release

Project description:The aim of the project is to identify the genes in megakaryocytes whose expression is modified upon 3D methylcellulose hydrogel culture compared to liquid culture. Megakaryocytes are the precursor cells of platelets. We previously showed that growing megakaryocytes in 3D hydrogel strongly modifies their differentiation and their morphology, being closer to the one in situ. We were thus interested into identifying these mechanobiological pathways induced by the confinement. We found that 3D culture notably upregulated genes related to the Rho GTPase pathway.
