Gene expression profile of HeLa cells overexpressing GFP tagged Nup88

ABSTRACT: Elevated expression of Nup88 is often observed in various cancers and correlates with poor prognosis. However, the underlying molecular mechanism by which Nup88 induces cancer malignancy remains unclear. Considering the possibility that this occurs due to changes in gene expression, we established HeLa R19 cells that stably overexpress either GFP or GFP-tagged Nup88 and screened for genes whose expression changes in a Nup88-dependent manner. Consequently, we found a significant downregulation of KIF7, which encodes a regulator of the Hedgehog signaling pathway. The co-overexpression of Kif7 in HeLa stable cell line overexpressing Nup88 suppressed Nup88-dependent migration activity, suggesting that the downregulation of KIF7 contributes to Nup88-induced cancer malignancy/cell migration.
