TFIIIB roadblocks control alternative cleavage and polyadenylation site selection

ABSTRACT: To test the functional impact of Pol III on controlling alternative CPA site selection, we depleted, Rna15p, a component of the cleavage and polyadenylation factor I, Bdp1p, a core component of TFIIIB, and Bdp1p/Rna15p together, from the nucleus and assessed the effect on 3´ UTR lengths of genes nearby tRNAs. Many genes show changes in 3’ ends after Bdp1p depletion indicating that transcripts can transcription termination can be due to TFIIIB roadblocks rather than transcript cleavage by cleavage and polyadenylation factors.
