Project description:Rheumatoid arthritis is a systemic autoimmune disease, but disease flares typically affect only a subset of joints, distributed in a pattern that is distinctive for each patient. Pursuing this intriguing pattern, we show that arthritis recurrence is mediated by long-lived synovial resident memory T cells (TRM). In three murine models, CD8+ cells bearing TRM markers remain in previously inflamed joints during remission. These cells are bona fide TRM, exhibiting failure to migrate from joint to joint, preferential uptake of fatty acids, and long-term residency. Disease flares result from TRM activation by antigen, leading to CCL5-mediated recruitment of circulating effector cells. Correspondingly, TRM depletion ameliorates recurrence in a site-specific manner. Human rheumatoid arthritis joint tissues contain a comparable CD8+-predominant TRM population, most evident in late-stage leukocyte-poor synovium, exhibiting limited T cell receptor diversity and a pro-inflammatory transcriptomic signature. Together, these findings establish synovial TRM cells as a targetable mediator of disease chronicity in autoimmune arthritis.

Project description:Objective: Chronic periodontitis typically affects individuals aged 40–50. Aging impairs immunity, sustains chronic inflammation, and enhances autoimmunity, a process called "immunosenescence", which contributes to the development of type 2 diabetes and rheumatoid arthritis (RA). Thymic involution after puberty reduces naive T cells, promoting senescent CD4+ T cells. The reason is that senescent T cells attempt to complement this reduction by proliferating to their limits, maintaining their absolute numbers through “homeostatic proliferation”. These cells, characterized by PD-1/CD153 expression, show reduced proliferation and increased senescence-associated secretory phenotype (SASP) cytokines. This study investigates how ligature-induced periodontitis (LIP) in mice affects CD4+ T cell senescence. Materials and Methods: Balb/c mice aged 5, 10, 18, 26, 34, and 42 weeks received LIP by silk ligation around maxillary second molars. Splenic CD4+ T cells were extracted and cultured with IL-2, anti-TCRβ, and anti-CD28 for 1 to 3 days to mimic homeostatic proliferation. Senescence markers were compared between LIP and control groups before and after in vitro stimulation. RNA sequencing (RNA-seq) analysis was performed to identify differentially expressed genes. Results: No significant difference was observed in PD-1/CD153 double-positive cells between the LIP group and the control group at any age of mice. However, after in vitro stimulation, the LIP group showed a higher proportion of PD-1/CD153 double-positive cells, peaking at 18 weeks. SASP cytokines (osteopontin, IL-6) in the LIP group were higher than that in the control graoup. Senescence-associated β-galactosidase (SAβ-gal)-positive CD4+ T cells were more prevalent, indicating accelerated senescence in LIP group. Additionally, fewer cells in LIP group were in the S phase, suggesting cell cycle arrest. RNA-seq suggested "inflammatory response," "T cell differentiation," “STAT protein phosphorylation," and “JAK-STAT signalling pathway" in the LIP group. Among the upregulated genes in the LIP group, Cx3cr1, a marker selectively expressed by senescent CD4⁺ T cells; Il1rn, which reflects the inflammation-regulating capacity of senescent CD4⁺ T cells by antagonizing IL-1β signalling; Il4, indicative of Th2 differentiation and B cell activation via the JAK/STAT6 pathway; and Endog, a mitochondrial nuclease implicated in age-related cellular stress and telomere dysfunction, were ranked high. Conclusions: These results suggest that LIP accelerates CD4+ T cell senescence, potentially exacerbating systemic inflammation. Future experiments will involve transferring splenic CD4+ T cells from LIP and control groups into T cell-deficient mice to evaluate in vivo homeostatic proliferation and its role in experimental RA.

Project description:Atherosclerosis is modulated by immune cells. Understanding the diversity and heterogeneity in the aorta in steady state, as well as during different stages of disease will help to determine mechanisms promoting disease progression. To characterize the immune cells, we performed single cell RNA sequencing of aortic CD45+ leukocytes from C57BL/6 (WT) on chow diet and early disease Ldlr-/- mice fed high fat diet (HFD) for 21-days. The single cell RNA-seq analyses revealed the heterogeneity of aortic macrophages in steady state and early atherosclerotic disease.

After one week of basal diet, rats belonging to the groups DEN and DEN+CLO underwent intraperitoneal injection of DEN (30mg/kg body weight) dissolved in NaCl 9â°. The two other groups were injected with NaCl 9â° alone. At 30mg/kg, DEN is non-necrogenic thus only exhibiting initiating properties. Twelve days after injection, diet from rats belonging to the groups CLO and DEN+CLO was changed for diet containing 5000ppm CLO for up to 608 days. Series of 5 rats from each group were necropsied at days 18, 46, 102, 264, 377, 447 (reverse phase from day 377: no more CLO in the diet for rats belonging to the DEN+CLO group), and for the CLO and Control group 524, and 608 days after the injection of DEN or saline. From day 524, half of the CLO-treated rats were kept on basal diet (reverse phase) until day 608.

Project description:Emerging evidence indicates that gingival-resident helper CD4+ T cells are major drivers of periodontal inflammation in response to commensal and pathogenic oral microorganisms. Whether tissue-resident memory CD8+ T cells (TRM), which principally safeguard against viruses and cancer but also drive certain autoimmune and inflammatory conditions, impact periodontitis progression and severity remain unknown. We asked whether local reactivation of oral CD8+ TRM of a defined antigen specificity could exacerbate ligature-induced periodontitis (LIP), a well-established model of periodontal disease in mice. Topical application of virus-mimicking peptides to the oral mucosa concurrent with LIP 1) intensified alveolar bone loss, 2) amplified gingival and cervical lymph node inflammation, and 3) stimulated gingival transcriptional changes in genes related to innate immune sensing and cell-mediated cytotoxicity. Therapeutic depletion of CD103-expressing oral CD8+ TRM in advance of LIP prevented exacerbation of disease. These observations provide evidence that oral CD103+ CD8+ TRM have the potential to participate in gingival inflammation, alveolar bone loss, and periodontitis.
