ABSTRACT: Sonic hedgehog-medulloblastoma (SHH-MB) represents a biologically diverse and clinically challenging subgroup, especially in high-risk variants characterized by metastatic dissemination. Identifying novel oncogenic mechanisms that can be therapeutically exploited is therefore a critical priority. Here, we uncover Pentraxin-3 (PTX3) as a previously unrecognized, SHH-enriched oncogenic mediator that promotes proliferation, migration, clonogenicity, and angiogenesis. Mechanistically, we demonstrate that PTX3 activates a TLR4/IRAK1/PI3K-Akt/GSK-3/β-catenin signaling cascade, establishing a key regulatory axis essential for SHH-MB growth. We used microarrays to detail the global programme of gene expression in DAOY shNT and shPTX3 cells.
