ABSTRACT: Temozolomide (TMZ) induces DNA alkylation lesions repaired primarily through base excision repair. To characterize transcriptional responses to TMZ in the context of impaired APE1 endonuclease activity, RNA-seq was performed on Pa03C Cas9 and E96A mutant cell lines following treatment. Cells were treated with TMZ for 4 hours or 8 hours, along with untreated controls. This dataset captures genotype-specific and time-dependent transcriptional responses to alkylation stress in PDAC cells.
