Biocompatibility results for endovascular medical device

ABSTRACT: In this project, we developed an endovascular medical device. To evaluate its biocompatibility, the device (experimental) or a clinical medical device (control) was co-cultured with human immune cells. We further conducted single-cell RNA sequencing to examine potential immunogenic responses at the cellular level.

Project description:We have developed an endovascular catheter-based device for sampling of the endothelium, intended for RNA-seq analysis. With an outer diameter of 0.23 mm it is capable of reaching peripheral vessels for endothelial sampling, and favourably compares to other approaches with stent retrievers or similar tools. Here, we have collected samples from liver and kidney vessels from swine and performed RNA-sequencing to validate the sample quality.

Effect of endovascular treatment on gene expression in patients with Budd-Chiari syndrome

Project description:Budd-Chiari syndrome (BCS) results in sinusoidal congestion and hepatocyte apoptosis, which can further progress to liver fibrosis, cirrhosis, and hepatocellular carcinoma. Endovascular (EV) treatment has been the primary therapeutic method and achieved excellent outcomes. However, whether EV treatment could reverse liver cirrhosis in patients with BCS is still unclear. To investigate the effect of endovascular treatment on liver cirrhosis in patients with Budd-Chiari syndrome, we performed gene expression profiling analysis of the liver from patients with or without EV treatment.

Project description:This study assess the biocompatibility of ZrO2 vs Y-TZP alloys on Saos-2 cells, through the combination of conventional toxicological assay, morphological observations and transcriptomic analysis performed on Saos-2 cells. The biocompatibility of ZrO2 alloys was evaluated in comparison with Y-TZP alloys, studying cell proliferation (WST-1) and cell morphology/adhesion (SEM) on human Saos-2 cell lines. Finally, an evaluation of gene expression was performed on Y-TZP alloys rough surface that seems more interesting regarding biocompatibility results.
