RNA-sequencing of cells responding to transfected RNA in unmodified, 5methoxyuracil and N1 methylpsuedouridine modified RNA.

ABSTRACT: HepG2 cells were transfected with unmodified, m1Ψ100-modified, and 5moU25-modified mRNA encoding a eGFP reporter and subjected to transcriptome sequencing after 24 hours. Over 3,500 differentially expressed genes (DEGs) were identified in response to unmodified mRNA with an absolute log2foldchange greater than 1 and an adjusted p-value <0.01, of these, 2316 genes were upregulated. Go term enrichment analysis on upregulated genes identified numerous regulated pathways, including expected terms for interferon and interleukin production, as well as regulation of innate immune response and defence against viruses. Gene set enrichment analysis against Reactome terms also identified characteristic activated pathways included TLR3 cascade response and RIG-I/MDA5 mediated responses, whilst inhibited pathways included those involved in translation.
