Project description:We obtained human embryonic and fetal lungs from 5-22 pcw for scRNAseq and scATACseq analysis. To focus on epithelial differentiation and region specialization, we deeply sampled 15, 18, 20 and 22 pcw lungs and separated proximal and distal regions while leaving lungs at 5, 6, 9 and 11 pcw intact. These cell samples (except for one at 6pcw) were split and processed for both scRNAseq and scATACseq.

Project description:Cannabis use during pregnancy is associated with adverse neurodevelopmental outcomes. However, the underlying mechanisms and effects of cannabis on fetal development are relatively unknown. We determined the impact of chronic delta-9-tetrahydrocannabinol (THC, cannabis’s main psychoactive component) exposure on fetal development in a rhesus macaque model using advanced imaging combined with molecular and tissue studies. Animals were divided into control (n=5) or THC-exposed (n=5) groups, which received a daily THC edible pre-conception and throughout pregnancy. Animals underwent fetal brain T2-weighted MRI at gestational days 85 (G85), G110, G135 and G155 (term is ~G168) to assess brain development. All animals underwent cesarean delivery with fetal cerebral spinal fluid (CSF) and brain collection at G155 for microRNA (miRNA) and histologic analysis. THC exposure did not demonstrate any fetal brain volumetric changes. However, histological findings in THC-exposed brains suggested brain dysregulation that may have long-term developmental implications. We identified two extracellular-vesicle-associated miRNAs (mir-448 and mir-199a-3p) with>2-fold change in CSF with maternal THC use. Amniotic fluid (AF) was collected at the 4 gestational ages (GA) and relative protein abundance changes measured with isobaric labeling quantitative proteomics. Changes in AF were observed across the GAs and in response to THC-treatment.

Project description:Autoimmune hepatitis (AIH) is an immune-mediated liver disease characterized by an imbalance between effector and regulatory immune responses. Epigenetic regulation, particularly via microRNAs (miRNAs), is emerging as a critical modulator of the immune system during inflammatory diseases. Cannabinoids are increasingly recognized for their immunosuppressive and anti-inflammatory properties; however, the impact of delta-8-tetrahydrocannabinol (Δ⁸-THC), a novel minor compound found in Cannabis plant with less psychoactive property than Δ⁹-THC, remains entirely unexplored in combating autoimmune hepatitis. To address this gap, we investigated the immunoregulatory and hepatoprotective effects of Δ⁸-THC in Concanavalin A (ConA)-induced murine model of AIH. We found that Δ⁸-THC effectively ameliorated ConA-induced liver injury by restoring intrahepatic innate and adaptive immune homeostasis through the preservation of tolerogenic Kupffer cells, suppression of inflammatory monocytes, neutrophils, and natural killer cells. Moreover, Δ⁸-THC reversed the inflammatory lymphoid microenvironment in the liver caused by ConA by decreasing inflammatory CD4+ T cells and promoting expansion of regulatory T cells. These effects were correlated with the preservation of hepatoprotective miR-199a-3p and miR-100-5p expression in hepatic immune cells that were markedly downregulated in ConA-treated mice. Ingenuity Pathway Analysis identified mTOR, a key immune regulator of T cell differentiation, as a target of these miRNAs. Consistently, Δ⁸-THC treatment reduced hepatic mTOR expression in vivo and in vitro. Functional assays using miRNA mimics and inhibitors confirmed direct regulation of mTOR by these miRNAs, where miRNA-mediated mTOR suppression promoted regulatory T cells differentiation and inhibited Th1 and Th17 responses. Together, these findings identify Δ⁸-THC is as a novel anti-inflammatory and hepatoprotective compound that attenuates autoimmune liver injury through epigenetic regulation of the miRNA-mTOR signaling axis, revealing its potential immunomodulatory effects in autoimmune and inflammatory diseases.

Project description:Cannabis is commonly used amongst reproductive age males. Our group and others have shown that chronic delta-9-tetrahydrocannabinol (THC) use adversely impacts male fertility, but less is known about the potential reversibility of these changes. Our study’s objective was to determine if THC discontinuation mitigates THC-associated changes in male reproductive health using a rhesus macaque (RM) model of daily THC edible consumption over a 280-day period (4 spermatogenic cycles). Testicular volume, serum male hormones, semen parameters, sperm DNA fragmentation, seminal fluid proteomics, and whole genome bisulfite sequencing (WGBS) of sperm DNA were assessed at pre-THC, moderate-THC, and heavy-THC dosing, and at 70 and 140 days after THC discontinuation. Chronic THC use resulted in significant testicular atrophy, increased gonadotropins, decreased serum sex steroids, and increased DNA fragmentation. THC discontinuation led to partial recovery of testicular volume, sex steroids and sperm DNA integrity. Seminal fluid proteome analysis revealed differential expression of proteins enriched for processes related to cellular secretion, immune response, and fibrinolysis. WGBS identified significant differential methylation in heavy-THC versus pre-THC sperm, with partial restoration of methylation after discontinuation of THC. Genes associated with differentially methylated regions (DMRs) were enriched for pathways involved in nervous system development and function. This is the first study demonstrating that chronic THC use in RMs adversely impacts male reproductive health, methylation of genes involved in development, and expression of proteins important for male fertility. THC discontinuation improves impacts to male fertility, including partial restoration of THC-associated sperm DMRs in genes important for development.

Project description:Cannabis is commonly used in pregnancy for symptoms of nausea and pain, especially in the first trimester. Cannabis remains a federally illicit drug in the United States, but local legalization trends have resulted in increased availability and a decreased perception of harm. Unfortunately, limited scientifically rigorous information exists to inform decisions on use. Delta-9-tetrahydrocannabinol (THC, the main psychoactive component of cannabis) can cross the placenta and bind to cannabinoid receptor 1 (CB1) on the fetus. Because CB1 receptors are expressed in cardiomyocytes and endothelial cells, this suggests that THC exposure may impact fetal cardiovascular development. To understand this impact, our group used an established rhesus macaque model of chronic edible THC use during pregnancy. Animals were slowly titrated to a heavy THC dose (2.5mg/7kg/day) for 4 months preconception. Dams continued this daily THC dose throughout pregnancy with c-section delivery near term. Our model showed a decreased heart-to-body weight ratio from THC exposure. We examined the underlying changes in this selective fetal growth restriction of the cardiovascular system through tissue, protein, and gene expression analyses. H istological analysis of coronal sections of the heart and cross sections of the aorta showed no changes in collagen expression or maturity. Western blot analysis of collagen III expression showed no changes in left or right ventricle. Elastin expression was also unchanged in the aorta. To assess transcriptional changes, we performed bulk RNA-sequencing of vascular cells in the fetal aorta, umbilical vein, and umbilical artery, which revealed differentially expressed genes involved in metabolism and inflammation. These results suggest potential cardiovascular harm from in utero THC exposure by altering endothelial metabolism and inflammation.

Project description:The study describes miRNA expression in colon tissue following delta 9 tetrahydrocannabinol (Δ9-THC) administration to chronically SIV-infected rhesus macaques. To identify the underlying molecular mechanisms underlying its anti-inflammatory effects, we simultaneously profiled miRNA and mRNA expression in colon of chronically simian immunodeficiency virus (SIV)-infected rhesus macaques (RMs) administered either vehicle (VEH/SIV; n=9) or Δ9- tetrahydrocannabinol (THC; THC/SIV; n=8). Relative to controls, differentially expressed miRNAs were ~2 fold higher in VEH/SIV than THC/SIV RMs. Proinflammatory miR-130a, miR-222 and miR-29b, Lipopolysaccharide-responsive miR-146b-5p and SIV-induced miR-190b were significantly upregulated in VEH/SIV RMs. Compared to VEH/SIV RMs, 10 miRNAs were significantly upregulated in THC-SIV RMs, among which miR-204 was confirmed to directly target MMP8, an extracellular matrix-degrading collagenase that was significantly downregulated in THC/SIV RMs. Moreover, THC/SIV RMs failed to upregulate proinflammatory miR-21, miR-141 and miR-222 and alpha/beta defensins, suggesting attenuated intestinal inflammation. Further, THC/SIV RMs showed higher expression of tight junction proteins (occludin, claudin-3), anti-inflammatory MUC13, keratin-8 (stress protection), PROM1 (epithelial proliferation) and anti-HIV CCL5. Trichrome mason staining detected significant collagen deposition (fibrosis) in the paracortex and B cell follicular zones of axillary lymph nodes from all VEH/SIV but none of the THC/SIV RMs, thus demonstrating the ability of THC to prevent lymph node fibrosis, a serious irreversible consequence of HIV induced chronic inflammation. Furthermore, using flow cytometry, we showed that THC suppressed intestinal T cell proliferation/activation (Ki67/HLADR) and exhaustion (PD1) and increased the percentages of anti-inflammatory CD163+ macrophages. Finally, while THC did not affect CD4+ T cell levels, it significantly reduced CD8+ T cell percentages in blood at 150 and 180 days post SIV infection. These translational findings strongly support a role for differential miRNA/gene induction and T cell activation in THC-mediated suppression of intestinal inflammation in HIV/SIV and potentially other chronic inflammatory diseases of the intestine.

Project description:Given the increasing use patterns of cannabis in the US, there is an urgent need to better understand the drug’s effects on central signaling mechanisms. Extracellular vesicles (EVs) have been identified as intercellular signaling mediators that contain a variety of cargo, including proteins. Here, we examined whether the main psychoactive component in cannabis, Δ9-tetrahydrocannabinol (THC), alters EV protein signaling dynamics in the brain. We first conducted in vitro studies which found that THC activates signaling in choroid plexus epithelial cells, resulting in transcriptional upregulation of the cannabinoid 1 receptor and immediate early gene c-fos, in addition to the release of EVs containing RNA cargo. Next, male and female rats were examined for the effects of either acute or chronic exposure to vaped THC on circulating brain EVs. Cerebrospinal fluid was extracted from the brain, and EVs were isolated and processed with label-free quantitative proteomic analyses via high-resolution tandem mass spectrometry. Interestingly, circulating EV-localized proteins were differentially expressed based on acute or chronic THC exposure in a sex-specific manner. Taken together, these findings reveal that THC acts in the brain to differentially modulate circulating EV signaling, thereby providing a novel understanding of how exogenous factors can regulate intercellular communication in the brain.

Project description:Delta-9-tetrahydrocannabinol (THC) is the primary psychoactive compound of the cannabis plant and an exogenous ligand of the endocannabinoid system. In previous studies, we demonstrated that a single microdose of THC (0.002mg/kg, 3–4 orders of magnitude lower than the standard dose for rodents) exerts distinct, long-term neuroprotection in model mice subjected to acute neurological insults. When administered to old, healthy mice, the THC microdose induced remarkable long-lasting (weeks) improvement in a wide range of cognitive functions, including significant morphological and biochemical brain alterations. To elucidate the mechanisms underlying these effects, we analyzed the gene expression of hippocampal samples from the model mice. Samples taken 5 days after THC treatment showed significant differential expression of genes associated with neurogenesis and brain development. In samples taken 5 weeks after treatment, the transcriptional signature was shifted to that of neuronal differentiation and survival. This study demonstrated the use of hippocampal transcriptome profiling in uncovering the molecular basis of the atypical, anti- aging effects of THC microdose treatment in old mice.
