A subset of pro-inflammatory spiny mice macrophages promotes a regenerative phenotype in mouse fibroblasts

Project description:Macrophages play an essential role in tissue regeneration. However, the ability to dissect the role of macrophages in regeneration from their role in wound healing with scar has been hampered by a lack of comparative systems. In this study, we use a mammalian model of tissue regeneration and scar formation to contrast the role of macrophages in both wound healing paradigms. The African Spiny mouse (A. cahirinus) can regenerate tissue of the external ear pinnae after 4mm biopsy punch. The common lab mouse (M. musculus) forms a scar after the same injury. We test the potential of bone marrow derived macrophages from both species to activate local ear fibroblasts and find macrophages from A. cahirinus are able to induce a matrix turnover phenotype in fibroblasts from both species. We identify growth factors and cytokines specific to macrophages derived from A. cahirinus, and using single cell RNAseq and screening with these factors, we identify populations of macrophages unique to a regenerating injury compared to scar forming injury. Finally we test how macrophage populations change over time in a regenerating injury and how they differ from each stage in a scar forming system

Transcriptomic characterisation of transitioning cell types in the skin of Atlantic salmon

Project description:Background The skin maintains the body’s integrity and serves as the first line of defence against pathogens, stressors and mechanical injuries. Despite the global significance of salmon in aquaculture, how cells transcriptomic profile varies during wound healing, remains unexplored. Teleost contain adult pluripotent cells that differentiate into various tissues, including bone, cartilage, tendon, ligament, adipose, dermis, muscle and connective tissue within the skin. These cells are pivotal for preserving the integrity of skin tissue throughout an organism´s lifespan and actively participate in the wound healing processes. In this study, we characterise the transcriptomic profiles of putative mesenchymal stromal cells (fibroblast-like adult stem cells) in healthy Atlantic salmon tissue and during the wound healing process. Results Single-nucleus sequencing and spatial transcriptomics were used to detect transcriptomic changes occurring during wound healing that are commonly associated with mesenchymal stromal cells. We followed the transcriptomic activity of these cells during an in vivo wound healing time course study showing that these cells become more transcriptionally active during the remodelling stage of wound healing. The changes detected give insights into the potential differentiation pathways leading to osteogenic and fibroblast lineages in the skin of Atlantic salmon. Conclusions We chart the transcriptomic activity of sub-clusters of putative differentiating stromal cells during the process of wound healing for the first time, revealing different spatial niches of the various putative MSC subclusters, and setting the stage for further investigation of the manipulation of transitioning cell types to improve fish health. Symmetry breaking of tissue mechanics in wound induced hair follicle regeneration [bulk RNA-seq]

Project description:Tissue regeneration is a process that recapitulates the molecular and mechanical aspects of development and evolution. We use the wound-induced hair neogenesis (WIHN) model to investigate the mechanical and molecular responses of the laboratory (Mus) and African spiny (Acomys) mice. Laboratory and spiny mice showed an opposite trend of spatiotemporal morphogenetic field for WIHN during wound healing, and wound stiffness gradient across the whole wound bed predicated pattern of hair formation. Using bulk and single-cell RNA-seq analysis and K14-Cre-Twist1 transgenic mice, we identified the central role of the Twist1 pathway as the mediator of epidermal-dermal interaction and the emergence of periodic hair primordia. Lastly, we generated a Turing model with an underlying measure of stiffness to support a two-scale tissue mechanic model to explain the setup of a morphogenetic field from a wound bed (mm scale) or periodically arranged hair primordia from a morphogenetic field (μm scale). Delineating the common and distinct chemo-mechanical events during regenerative wound healing between laboratory and African spiny mice reveal its evo-devo advantages, which provide new perspectives for regenerative medicine.

Single-Cell Transcriptomic Profiling of colonic wounds and intact tissues in Hgfac WT and KO mice

Project description:Wound healing is orchestrated by a spatial and temporal network of intercellular communication between epithelial cells, the stromal compartment, and the immune system. We found that Hgfac KO mice showed delayed wound healing in an endoscopic wound model. To dissect the celluar and molecular mechanisms of tissue healing, we collected tissues from day2 wounds or intact tissues from Hgfac WT and KO mice using a skin biopsy punch and dissociated cells for scRNA-seq analysis.

Characterization of wound healing in Atlantic salmon (Salmo salar) post-smolt, and the effect of stress on the healing process

Project description:The aim of this study was to assess the effect induced by high fish density on cutaneous wound healing in post-smolt Atlantic salmon. Our findings suggest that high fish density enhances inflammation and represses cell proliferation, tissue secretion and collagen synthesis in the healing wounds. These processes are further manifested in the different layers of the skin, such as delayed re-epithelialization, altered mucus response, reduced scale mineralization, enhanced wound pigmentation and delayed organization of dense connective tissue. Temporarily changes in wound healing mechanisms result in permanent differences in wound contraction. In conclusion, our results show that high fish density dispose the fish for an enhanced immune response and delayed tissue repair, which ultimately results in delayed wound healing.
