Project description:Global analysis of H3K4 methylation defines MLL family member targets and points to a role for MLL1-mediated H3K4 methylation in the regulation of transcriptional initiation by RNA polymerase II A common landmark of activated genes is the presence of trimethylation on lysine 4 of histone H3 (H3K4) at promoter regions. The Set1/COMPASS was the founding member and the only H3K4 methylases in S. cerevisiae, however, in mammals at least six H3K4 methylases Set1A/B and MLL1-4 are found in COMPASS-like complexes capable of methylating H3K4. To gain further insight into the different roles and functional targets for the H3K4 methylases, we have undertaken a genome-wide analysis of H3K4 methylation pattern in wild-type Mll1+/+ and Mll1-/- mouse fibroblasts (MEFs). We found that Mll1 is required for the H3K4 trimethylation of less than 5% of promoters carrying this modification. Many of these genes, which include developmental regulators such as Hox genes show decreased levels of RNA polymerase II recruitment and expression concomitant with the loss of H3K4 methylation. Although Mll1 is only required for the methylation of a subset of Hox genes, Menin, a component of the Mll1 and Mll2 complexes, is required for the overwhelming majority of H3K4 methylation at Hox loci. However, the loss of MLL3/4 and/or the Set1 complexes have little to no effect on the Hox loci H3K4 methylation or expression levels in these MEFs. Together these data provide insight into redundancy and specialization of COMPASS-like complexes in mammals and provide evidence on a possible role for Mll1-mediated H3K4 methylation in the regulation of transcriptional initiation. Expression arrays were done with Mll1+/+ and Mll1-/- mouse embryonic fibroblasts. Four replicates were done (dyes were swapped). DNA was hybridized to Agilent Mouse Whole Genome Expression Arrays (4x44k).

Project description:Purpose: To characterize transcriptional changes associated with homozygous inactivation of Dot1l or Mll1 in MN1 driven AML Methods: We sequenced mRNA from murine LSK-cells transformed using forced expression of MN1 (MSCV-MN1-IRES-GFP), and transduced with Cre-vector to inactivate either Dot1l or Mll1. Cells were sorted for Cre-expression (pTomato fluorescent marker) or expression of an inert control vector. Results: Inactivation of either Dot1l or Mll1 in this model leads to a substantial delay or complete abrogation of leukemia development.Loss of Dot1l or Mll1 are associated with gene expression changes that have substantial overlap. In addition, genes that are downregulated follwing inactivation of Dot1l or Mll1 have substantial overlap with the gene set upregulated in MN1 transduced CMPs. Conclusions: MN1 mediated leukemogenesis is associated with a gene expression program that dependes on Mll1 and Dot1l Examination of mRNA levels between Dot1l f/f and Dot1l ko, and Mll1 f/f and Mll1 ko.

Project description:The interaction between MLL1 And Menin controls cancer cell proliferation, migration, and tumor progression. MLL1 depletion reduced 3D cell migration in vitro and led to lesser metastatic burden and prolonged survival in vivo. Mechanistically, MLL1-Menin interaction controls actin filament assembly and protrusion generation through IL-6-pSTAT3-Arp3 axis. MLL1-Menin interaction also controls TGF-β1-mediated acto-myosin contractility via Gli2-ROCK1/2-pMLC2 axis. Additionally, MLL1-menin inhibition decreased cell proliferation via a multitude of cell-cycle related pathways. Mice bearing MLL1-depleted tumors exhibited decreased significant lung metastatic burden when sacrificed at a set tumor size, indicating that MLL1 played a role in the invasion of tumor cells in vivo. Finally, MLL1-Menin inhibition was additive with standard chemotherapeutics, both in vitro and in vivo.

Project description:The trithorax H3K4 histone methyltransferase (HMT) MLL1 has important roles for early embryonic development, hematopoiesis and neurogenesis through regulation of Hox and homeodomain factor expression. MLL1 has been previously implicated in activation of Myf5 expression through an interaction with Pax7. Here, we find that in vivo, MLL1 is necessary for efficient muscle regeneration, and for maintenance of muscle stem and progenitor cells. Loss of MLL1 in cultured myoblasts reveals an essential role for proliferation and expression of both Myf5 and Pax7. Loss of Myf5 is conditional on loss of Pax7 and exogenous Pax7 rescues Myf5 in the absence of MLL1 suggesting a role for MLL1 in Pax7 expression but not Pax7 activity. Importantly, Mll1 knockout results in a minor decrease to H3K4me3 at Pax7, a 40% decrease in Pax7 mRNA and an 85% decrease in Pax7 protein, suggesting a previously proposed non-HMT role for MLL1 may involve linking transcription to translation.

Project description:By removing MLL1 from mouse retinal progenitors, we discovered that MLL1 plays multiplex roles in retinal development by regulating: 1) progenitor cell proliferation and cell cycle progression; 2) retinal cell composition; 3) maintenance of horizontal neurons; 4) formation of functional synapses between neuronal layers; and 5) visual function development. Altogether, our results suggest that MLL1 is an indispensable regulator for the development of retinal structure and function.

Project description:T follicular helper (TFH) cells are essential for developing protective antibody responses following vaccination. Greater understanding of the genetic program leading to TFH differentiation is needed. Chromatin modifications are central in the control of gene expression. However, detailed knowledge of how chromatin regulators (CRs) regulate differentiation of TFH cells is limited. We screened a large shRNA library targeting all known CRs in mouse and identified the histone methyltransferase Mll1 as a positive regulator of TFH differentiation. Loss of Mll1 expression reduced formation of TFH cells following acute viral infection or protein immunization. In addition, expression of the TFH lineage-defining transcription factor Bcl6 was reduced in the absence of Mll1. Transcriptomics analysis identified Lef1 and Tcf7 as genes dependent on Mll1 for their expression, which provides one mechanism for the regulation of TFH differentiation by Mll1. Taken together, CRs such as Mll1 substantially influence TFH differentiation.
