Splenic CD8+ and CD8- DCs

Project description:Analysis of gene expressions in mouse splenic dendritic cells (DCs). DCs were purified into two subsets, CD8-positive and -negative ones. DCs were expanded in vivo by injecting Flt3L-producing tumors into the backs of C57BL/6 mice. To expand DCs in vivo, Flt3L-producing B16 melanoma cells were injected to the backs of mice. Then, 10-12 days later, splenic DCs were enriched by MACS and purified into CD3-B220-CD8a+CD11c+ and CD3-B220-CD8a-CD11c+ cells by FACS cell sorter.

HLA-E Presents Glycopeptides from the Mtb Protein MPT32 to Human CD8 T Cells

Project description:Infection with Mycobacterium tuberculosis (Mtb), the bacterium that causes Tuberculosis, remains a global health concern. Both classically and non-classically restricted cytotoxic CD8+ T cells are important to the control of Mtb infection. We and others have demonstrated that the non-classical MHC I molecule HLA-E can present pathogen-derived peptides to CD8+ T cells. In this manuscript, we identified the antigen recognized by an HLA-E-restricted CD8+ T cell clone isolated from an Mtb latently infected individual as a peptide from the Mtb protein, MPT32. Recognition by the CD8+ T cell clone required N-terminal O-linked mannosylation of MPT32 by a mannosyltransferase encoded by the Rv1002c gene. This is the first description of a post-translationally modified Mtb-derived protein antigen presented in the context of an HLA-E specific CD8+ T cell immune response. The identification of an immune response that targets a unique mycobacterial modification is novel and has practical impact in the development of vaccines and diagnostics.

Aeromonas spp MTB Infection May Causally Link to Ulcerative Colitis

Project description:Ulcerative colitis (UC) is an inflammatory bowel disease characterized by disruption of the epithelial barrier, and its etiology has long been enigmatic. We here observed a significant depletion of macrophages directly beneath the UC epithelial layer, and hypothesized that this depletion may be caused by a toxic bacteria. By screening fecal bacteria of UC patients, we identified the causative bacteria as Aeromonas spp MTB (macrophage-toxic bacteria), a novel variant of Aeromonas genus. The expressed aerolysin was preferentially toxic towards macrophages, and severs as a primary virulent factor. MTB efficiently colonized mice after pretreatment with DSS and antibiotics, and thereby induced UC-like colitis. Moreover, both aerolysin toxin and fecal MTB respectively distributed in UC colon tissue and stool with high frequency. We thus proposed that MTB infection was causally linked to ulcerative colitis. As aerolysin antibody mitigated the colitis phenotypes, our result also revealed an innovative therapeutic approach for UC.
