Expression data from rat heart tissue ischemia/reperfusion (I/R) treated for 12,24 hours

ABSTRACT: Acute pancreatitis (AP), a common inflammatory disorder, can lead to systemic complications such as myocardial injury. However, the dynamic transcriptomic changes and key molecular mechanisms behind these pancreatic-associated cardiac complications remain unclear. We investigated SAP-mediated gene expression alteration associated with the development of cardiac injury by microarray in a rat model.

CircRNA expression profiles of pancreatic tissues from severe acute pancreatitis mice

Project description:Severe acute pancreatitis (SAP) is the most serious type of pancreatitis with high morbidity and mortality. The underlying mechanism behind SAP pathogenesis is complex and remains elusive. In the present study, next-generation RNA sequencing was utilized to identify circRNA transcripts in the pancreatic tissues from three SAP mice and three matched normal tissues. Our results discovered that 56 circRNAs were differently expressed in SAP compared with normal control. Our findings may provide novel insights for pathophysiological mechanism of SAP and offer novel targets for SAP.

Hepatic transcriptome analysis of control, acute pancreatitis and severe acute pancreatitis mice

Project description:Purpose: The purpose of this study was to compare the hepatic transcriptome in the control group, acute pancreatitis, and severe acute pancreatitis, in order to identify metabolic gene changes during pancreatitis. Methods: 6-week Balb/c mice were subjected to AP (caerulein), SAP (caerulein and LPS) and control (saline), and mice liver tissues were harvested at 24 hours for RNA preparation (n=8 each group). Results: Among 11952 mapped genes, 8977 differentially expressed genes were identified by RNA-seq, including 351 genes induced in AP compare to Control and 5249 genes down regulated in SAP compare to AP. The genes up regulated were mainly related to fatty acid oxidation and ketone body synthesis especially Cpt2 and Acadvl, while genes that down regulated were associated with energy metabolism and inflammation. The overlap of two gene sets were 96 genes that involved in fatty acid metabolism. Conclusions: Our study revealed that in AP, genes associated with ketone body synthesis are upregulated in liver, in SAP, fatty acid oxidation related genes are downregulated in liver. Our data revealed the crosstalk between pancreas and liver, illustrated the difference of metabolism in AP and SAP.

Comprehensive analysis of differentially expressed lncRNA, and mRNA and their ceRNA networks in mice with the initial stage acute pancreatitis

Project description:Acute pancreatitis (AP) is a prevalent digestive disorder with significant morbidity and mortality rates. This study, for the first time, employed high-throughput sequencing on pancreatic tissues from normal and AP mice to identify differentially expressed long noncoding RNAs (lncRNAs) and mRNAs, as well as related signaling pathways, in the early stage of AP. We found 261 lncRNAs and 1,522 mRNAs to be differentially expressed, with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses revealing gene functions enriched in cellular metabolism, intercellular structure, and positive regulation of inflammation. By constructing lncRNA-miRNA-mRNA networks, we explored underlying mechanisms and interactions in AP. Notably, knockout of Neat1 in mice inhibited pyroptosis in AP and severe acute pancreatitis (SAP) models, suggesting Neat1's pivotal role in AP pathogenesis. These findings highlight the importance of lncRNAs, particularly Neat1, in AP, offering new insights into SAP pathogenesis and potential therapeutic targets.

Expression data from lung tissues of minipig with left lung ischemia-reperfusion

Project description:Lung ischemia-reperfusion (I/R) injury remains one of the common complications after various cardiopulmonary surgeries. I-R injury represents one potentially maladaptive response of the innate immune system which is featured by an exacerbated sterile inflammatory response triggered by tissue damage. Thus, understanding the key components and processes involved in sterile inflammation during lung I-R injury is critical to alter care and extend survival for patients with acute lung injury. We constructed a minipig surgical model of transient unilateral left pulmonary artery occlusion without bronchial involvement to create ventilated lung I-R injury. Lung tissues from minipig with sham operation (one sample), left side lung tissues (the operated side)(one sample) and right side lung tissues (the non-operated side)(one sample) from minipig with lung ischemia-reperfusion were submitted for gene expression array analysis.

RNA-seq analysis of young swine following cardiac ischemia/reperfusion injury.

Project description:This study was a part of a larger study assessing the response of young pigs to cardiac ischemia/reperfusion (IR) injury. A mild cardiac injury approach (IR) was used in post-weaned pigs (1-month), to assess regenerative repair in young large mammals after transient ischemic injury. Female and male postnatal day (P)30 pigs were subjected to cardiac ischemia (1-hour) by occlusion of the left anterior descending artery followed by reperfusion (IR), or to sham operation. In pigs subjected to IR, myocardial damage occurred, indicated by increased circulating cardiac troponin I 2-hours post-ischemia. In addition, cardiac ejection fraction (EF) was significantly decreased 2-hours post-ischemia and reduced EF was maintained to the 4-week study end-point. Histology demonstrated evidence of CM cell cycling at 2-months of age in multinucleated CMs in both sham-operated and IR pigs. Following IR, regional scar formation and inflammation in the epicardial region proximal to injury were observed 4-weeks post-IR. Sex differences in cardiac function and collagen deposition were found, highlighting the importance of representing both sexes in cardiac injury studies. Together, our results describe an effective novel cardiac injury model in 1-month old swine, at a time when CM are still cycling. However, pigs subjected to IR show a prolonged decrease in cardiac function, and the formation of a small, regional scar with increased inflammation. Together, these data demonstrate that 1-month old pigs do not regenerate myocardium, but form a scar, after transient IR injury.

Decorin protects cardiac myocytes against simulated ischemia/reperfusion injury

Project description:Primary cardiac myocytes isolated from neonatal Wistar rats were cultured and simulated ischemia/reperfusion injury were conducted on them in the presence or absence of decorin. Decorin is a small leucin-rich proteoglycan, which has a cardiocytoprotective effect. The underlaying mechanism of this cardiocytoprotective phenomenon is unknown, therefore our aim was to investigate the changes in the mRNA expression between the decorin (3nM) treated and vehicle-treated control cells.

Role of KLF5 in Ischemia Reperfusion Injury

Project description:Acute myocardial infarction remains a leading cause of morbidity and mortality worldwide. Our previous studies have investigated the role of the transcription factor Kruppel-like factor 5 (KLF5) in various cardiac disease models, including diabetic cardiomyopathy and ischemic heart failure, and have linked it to the regulation of cellular stress responses, particularly those associated with oxidative stress and metabolic dysregulation. This study examines the role of KLF5 in exacerbating myocardial ischemia/reperfusion (I/R) injury and explores the potential cardioprotective benefit of KLF5 inhibition. Using the surgical model of myocardial I/R injury in both pigs and mice, we observed that KLF5 expression is upregulated in cardiomyocytes during early reperfusion. This increase in KLF5 expression is accompanied by enhanced oxidative stress, apoptosis, fibrosis, and adverse cardiac remodeling. Pharmacologic and cardiomyocyte-specific genetic inhibition of KLF5 significantly reduced infarct size, oxidative stress, and cell apoptosis markers, while preserving cardiac function over both acute and long-term reperfusion periods.
