Piezo1 Modulates Hematopoietic Homeostasis and Chronic Myelomonocytic Leukemia Progression [low-put RNA-seq]

ABSTRACT: We demonstrated that PIEZO1 is essential for sustaining hematopoietic homeostasis. To elucidate the underlying mechanism of Piezo1 in normal hematopoiesis, we analyzed and compared the gene expression profiles of hematopoietic stem cells (HSCs) — which were isolated via flow cytometry from Piezo1 floxed/floxed; Vav-iCre mice and control mice. Our findings revealed that, among the differentially expressed genes (DEGs) in Piezo1 conditional knockout (cKO) HSCs, the cell adhesion pathway was significantly enriched and exhibited downregulation. This downregulation of the cell adhesion pathway was associated with reduced expression of integrin beta1.
