Selection of cord-blood donors for NK cell-based anti-tumor immunotherapy

ABSTRACT: Donor heterogeneity in cord blood?derived NK cells can influence the efficacy of NK cell?based anti-tumor immunotherapy. Here, we profile single-cell gene expression in ex vivo-expanded cord blood NK cells and primary cord blood to determine how baseline NK-cell subset composition shapes subsequent immune effector activity.

Gene expression profiling of human NK cells

Project description:In order to identify genes expression in different types of human NK cells, we sorted normal decidual NK cells during the first trimester, NK cells from peripheral blood and cord blood for analysis using the Agilent Whole Human Genome Microarray. Samples were collected from healthy adult donors after obtaining informed conset according to the Ethics Committee of the University of Science & Technology of China. CD56+CD3- NK cells have been sorted from normal decidua, cord blood and peripheral blood by FACS Aria. Then Whole Human Genome Microarray was employed as a discovery platform to identify genes differentially expressed in each types of NK cells.

microRNA expression signatures for human primary NK cells

Project description:We examined miRNA profiles of human NK cells from different cell compartments (peripheral blood, cord blood, and uterine deciduas) and of NKT and T cells from peripheral blood, and identified distinct classes of up-regulated microRNAs in different human NK cells. For the microarray assay, RNAs from 6 donors with equal amount were pooled together to get each cell sample.

Identification and characterization of human Natural Killer (NK) lineage restricted progenitors

Project description:Natural killer (NK) cells represent one of three lymphoid lineages and play a vital role in controlling viral infections and cancer. In contrast to B and T lymphopoiesis where cellular and regulatory pathways have been extensively characterized, the cellular stages of early human NK-cell commitment remain poorly understood Here we described a novel NK-lineage restricted progenitor (NKP) in fetal development, umbilical cord blood and adult bone marrow. We used microarrays to detail the global programme of gene expression genes of this new progenitor. CAR-NK cells derived from cord blood predominantly originated from CD56-CD7+CD34-HLA-DR- lineage markers- NK progenitor cells

Project description:Cord blood (CB)-derived chimeric antigen receptor (CAR)-natural killer (NK) cells targeting CD19 has been shown to be effective against B cell malignancies. While human CD56+ NK cells can be expanded in vitro, it is also known that NK cells can be differentiated from hematopoietic progenitor cells. It is still unclear whether CAR-NK cells are originated from mature NK cells or NK progenitor cells in CB. Here we found that CAR-NK cells are predominantly derived from the CD56- NK progenitor cells. We first found that substantial numbers of CD19 CAR-NK cells were produced from the CD56- CB mononuclear cells after in vitro culture for two weeks. Single cell RNAseq analysis of CD56-CD3-CD14-CD19- CB mononuclear cells revealed that those cells were subdivided into three subpopulations based on the expression of CD34 and HLA-DR. NK cells were predominantly produced from the CD34-HLA-DR- cells. In addition, in the CD34-HLA-DR- cells, only CD7+ cells could differentiate into NK cells. These results indicate that CD56-CD7+CD34- HLA-DR-lineage marker (Lin)- cells are the major origin of human CB-derived CAR-NK cells, indicating that we need to develop methods to enhance the quality and quantity of NK cells produced from these NK cell progenitor cells.

CAR NK cell production from human cord blood cells is enhanced by stimulation of the IL15 receptor on NK progenitor cells

Project description:Human cord blood (CB)-derived CAR NK cells have demonstrated significant anti-tumor efficacy in clinical trials. We recently reported that CB-derived CAR NK cells are mainly derived from CD7+CD56-CD34-HLA-DR-Lin- NK precursors. Here we show that CAR NK cell production from human CB cells can be enhanced by stimulation of the IL15 receptor on NK precursors. In CB CD56-CD34-HLADR-Lin- cells, the IL15 receptor was expressed exclusively on the CD7+ NK cell precursors but not on the CD7- population. The production of mature NK cells from the purified NK cell precursors or CD3- CB cells was significantly enhanced by using K562 feeder cells expressing membrane-bound (mb) IL15 in addition to 4-1BB ligand and mbIL21. The in vitro and in vivo anti-tumor efficacy of CAR NK cells generated using K562 feeder cells expressing 4-1BBL, mbIL15 and mbIL21 was comparable to that of CAR NK cells generated using the K562 feeder cells expressing only 4-1BBL and mbIL21. These results suggest that the use of K562 feeder cells expressing mbIL15 in addition to 4-1BBL and mbIL21 increases the yield of CAR NK cell production from CB cells, while the clinical efficacy and safety of the CAR NK cells generated should be tested in the future.
