RNA-seq of placentas from FNDC5-deficient and wild-type mice at gestational day 17.5

ABSTRACT: Fibronectin type III domain–containing protein 5 (FNDC5), the precursor of the myokine irisin, has been implicated in metabolic regulation and pregnancy outcomes, but its role in placental development is not well understood. To investigate how FNDC5 deficiency affects the placental transcriptome, we performed mRNA sequencing on placentas collected at gestational day 17.5 from FNDC5 knockout (FNDC5-/-) and wild-type mice. Total RNA was isolated from whole placentas, poly(A)+-selected, and used to generate strand-specific libraries with the NEBNext Ultra II Directional RNA Library Prep Kit, followed by paired-end sequencing on an Illumina NovaSeq 6000. These data enable identification of FNDC5-dependent changes in placental gene expression associated with alterations in fetal–placental growth and maternal body weight.
