Gene expression profiles at the single-nucleus level in heart tissue after myocardial infarction and saNppa-LNP treatment

ABSTRACT: This project aims to investigate the cellular and molecular mechanisms underlying the therapeutic effects of a self-amplifying RNA encoding atrial natriuretic peptide (saNppa) in myocardial infarction (MI). In this study, a myocardial infarction model was established in C57BL/6 mice by coronary artery ligation. On the day of surgery, mice received intramuscular injection of saNppa formulated in lipid nanoparticles (saNppa-LNP) or phosphate-buffered saline (PBS) as control. Single-nucleus RNA sequencing (snRNA-seq) was performed on left ventricular tissue collected at Day 7 post-MI and treatment. Three experimental groups were included: sham-operated mice, MI mice treated with PBS (MI+PBS), and MI mice treated with saNppa-LNP (MI+saNppa-LNP). This dataset provides a comprehensive single-nucleus transcriptional landscape of the post-infarct heart and enables cell type–specific analysis of transcriptional changes associated with myocardial infarction and saNppa self-amplifying RNA therapy.
