Project description:Background Both high fiber diet and N-Carbamylglutamate (NCG) individually contributes to improve reproductive efficiency in sow. However, the potential synergistic effects and the underlying mechanisms are not well-understood. Here, Landrace x Yorkshire sows were selected to investigate the synergistic effects of high fiber diet and 0.05% NCG on reproductive performance using metagenomic, metabolomic, and transcriptomic analysis.Results The study revealed that high fiber with NCG (H+N) significantly increased litter weight at weaning and the apparent nutrient digestibility. Metagenomic analysis revealed H+N increased the abundance of lactobacilli across multiple taxonomic levels (from order to species level, including Lactobacillus sp. 910589175), which was positively correlated with elevated levels of the lipid metabolites L-α-glycerylphosphorylcholine and taurocholic acid. Subsequent transcriptomic profiling indicated activation of the PI3K-Akt signaling pathway, which was associated with improved litter weight at weaning and the apparent nutrient digestibility.Conclusion The synergistic effect of high-fiber diet and NCG supplementation enhances reproductive efficiency in sows by modulating gut microbiota (specifically lactobacilli), increasing beneficial lipid metabolites, and activating the PI3K-Akt signaling pathway, thereby improving nutrient digestibility and litter weight.

Defining Cellular Diversity at the Swine Maternal-Fetal Interface Using Spatial Transcriptomics and Organoids [MFI017_Visium]

Project description:In this study we had two primary aims: 1.) spatially define the transcriptional signatures of porcine maternal-fetal interface and 2.) develop and validate an organoid model which better recapitulated the porcine placenta. Using mid-gestation maternal-fetal interfaces of commercial landrace/large white composite gilts we performed spatial transcriptomics (n=4 interfaces) using Visium v1 spatial transcriptomics. We then went on to isolate trophoblast organoids from fresh-term placentas from crossbred sows consisting655 of Yorkshire, Large White, and Landrace breeds. We then characterized the transcriptional profile of these organoids using bulk RNA-seq from 3 seperate lines using a standard Illumina library preparation. To characterize these organoids we performed single cell RNA-sequencing on 3 separate lines of swine trophoblast organoids using a standard 10x Genomics Single Cell 3' Gene Expression platform. All reads/samples were mapped to Sus scrofa v11.1.

Detection of differentially expressed genes in Day 30 embryos during gestation in primiparous (PP) sow with different intermittent suckling and breeding strategies

Project description:Transcriptional profiling of Day 30 embryos (D30E) was performed. First parity sows were submitted to an ovulation-induction protocol, intermittent suckling (IS), during lactation. IS consisted of 8 h/d separation from their litters during the last 7d of a 28d lactation. During separation, sows received boar exposure. There were 3 treatments: control (C28, n=19), where piglets were weaned at D28 of lactation and were bred after weaning and two IS treatments: sows were either bred at their first induced estrus during lactation (IS21FE, n=18), or were “skipped” and bred at their second estrus (IS21SE, n= 17) which occurred after final weaning at D28. Sows were slaughtered and embryos were collected on D30 of gestation for DNA PCR sexing. Later, D30E from the same sex with similar weight were pooled for further microarray investigation.

miRNA Expression in Early Porcine Pregnancy

Project description:This study examined the expression of pig-specific microRNAs (miRNAs) at gestation day 20 (gd20) of pregnancy in Yorkshire sows. Tissue differences in miRNA expression, and miRNA differences between healthy and arresting embryo attachment sites (i.e., healthy endometrium vs. arresting endometrium; healthy trophoblast vs. arresting trophoblast), were of prime interest. For more information, please refer to the primary research paper. Paired endometrium and trophoblast samples were collected at gestation day 20 from two conceptus attachement sites (1 healthy, 1 arresting) per sow (n=3). Endometrial samples were collected from four non-pregnant sows at mid-estrus.
