Cell-Specific DNA Methylation Markers in Plasma cfDNA Reveal Diagnostic Potential for Head and Neck Cancer


ABSTRACT: Head and neck cancer is the seventh most common type of cancer worldwide. This study screened cell-specific methylation sites using WGBS data of healthy tissues (GSE186458), identifying 1,157 sites, including 146 hypermethylated and 1,011 hypo-methylated sites. By clustering adjacent sites within 150 bp, 11 composite methylation markers were obtained, showing strong cell type specificity. Validation in plasma cfDNA revealed marker fragments in cancer patients but not in healthy individuals, supporting their diagnostic potential.
