LncRNA and mRNA expression characteristics and bioinformatics analysis of exosomes from sheep ovaries with different reproductive capacities

ABSTRACT: Hu sheep (HS) is known for its year-round estrus and multiple births, and is an ideal model for studying the mechanisms of high reproductive capacity in livestock. We established an RNA sequencing dataset to compare the expression profiles of long non coding RNA (lncRNA) and messenger RNA (mRNA) in the ovarian tissue exosomes of Hu sheep and low reproductive sheep breeds (Mongolian sheep) during estrus period.

A single-cell atlas of the single versus multiple parous Hu Sheep ovary

Project description:Follicular development is a highly coordinated process in Hu sheep. Follicle-cyclic recruitment, spatial displacement, follicle atresia, and ovulation are implicated events resulting from the somatic cells' release of molecular signals. Hu sheep is a high-quality sheep breed with high fecundity in China and is ideal for investigating high reproductive traits. In the current study, the sheep with lambing number ≥3 in three consecutive lambing records were assigned to the HLS group, and lambing number = 1 as the LLS group selected from the same farm with three consecutive lambings. Three randomly picked ewes were slaughtered within 12 h of estrus, and unilateral ovarian tissue was collected and analyzed by single-cell RNA sequencing in each group. A total of five types of somatic cells were identified, and corresponding expression profiles were mapped in the ovaries of the Hu sheep. Additionally, the results of the difference in ovary somatic cell expression profiles between HLS and LLS present that the differences between multiples vs. singleton Hu sheep were mainly clustered in the GCs. In addition, 4 granulosa cell subtypes were identified. GeneSwitches results revealed the opening of JPH1 expression and the closure of LOC101112291, which leads to different evolutionary directions of the granular cells. The expression levels of FTH1 and FTL in GCs of Hu sheep in the HLS group were significantly higher, which inhibited necroptosis and ferroptosis of mural-GCs from decreasing follicular atresia. This study constructed the cellular atlas of the ovary and revealed related biological characteristics at the cellular molecular level. It provides a theoretical basis for the mechanisms underlying the differences in ovulation numbers, which contributes to breeding high-fertility sheep and molecular genetics-based selection.

DIA Proteomics Unveils cAMP and HPT Axis in Hu Sheep Spermatogenesis

Project description:Reproductive efficiency in sheep is critically important for livestock production. This study provides the first multi-tissue proteomic characterization of high fecundity in Hu sheep, suggesting that their reproductive superiority may originate from hypothalamic-driven regulation, efficient coordination of key signaling pathways such as cAMP, and complex interactions among functionally important proteins across tissues. These findings offer valuable molecular insights into the regulation of reproductive traits in sheep, and the identified key proteins and pathways represent promising targets for further functional validation and molecular breeding applications.

The Analysis of Genes that Influence Sheep Follicular Development by Different Nutrition Levels during the Luteal Phase using Expression Profiling

Project description:Purpose: Nutrition is an important factor that regulates reproductive performance of sheep and affects follicle development. However, the correlation between nutrition and follicle development is poorly understood at molecular level. In order to study its possible molecular mechanisms, we performed expression profiling of granulosa cells isolated from sheep that were fed with different nutrition levels during luteal phase. Methods: To do this, ewes received a maintenance diet (M) and their estrus was synchronized by intravaginal progestogen sponges for 12 days. Ewes were randomly divided into the short-term dietary restricted group (R; 0.5ÃM) and nutrient-supplemented group (S; 1.5ÃM). RNA samples were extracted from granulosa cells. Transcriptome libraries from each group were constructed by Illumina Sequencing. Results: In 18,468 detected genes, 170 genes were significantly differentially expressed; of which, 140 genes were up-regulated and 30 genes were down-regulated in group S relative to group R. These genes could be candidates regulating follicular development in sheep. GO, KEGG and Clustering analyses were performed. Conclusions: These data in our study are abundant genomic recourse to expand our understanding of the molecular and cellular events underlying follicle development. Sixteen multiparous Hu sheep (3-4 years; 42.2 Â± 1.2 kg) were randomly assigned to two groups: the short-term nutrient-supplemented group (S; n=8), and the dietary restricted group (R; n=8).Expression profiling of granulosa cells were performed by Illumina sequencing platform (HiSeq 2000).

Scanning and Mining of High Fecundity Genes by Oxford Nanopore Technologies (ONT) in Sheep (Ovis aries) Pituitary

Project description:The pituitary gland is an important endocrine organ that regulates estrus and reproduction in sheep mainly through hormone synthesis and secretion. In present study, Small-tailed Han sheep (high-reproduction group, HP group), and Wadi sheep (lower-reproduction group, LP group), were used as the experiment materials, and the differential expressed genes (DEGs) were scanned and mined by Oxford Nanopore Technologies (ONT) in pituitary. A total of 7123 DEGs were found, including 3551 genes that were upregulated and 3572 genes that were downregulated in HP group. Go and KEGG related to pituitary function and reproduction were enriched, including reproductive processes, responses to stimuli, and synapses. mTOR signaling pathway, PI3K-Akt signaling pathway, cAMP signaling pathway, ERK1/2 signaling pathways and MAPK signaling pathways. The DEGs detected in this study were involved in the development of tissues and organs and the secretion of hormones in the endocrine system. These findings suggest that these genes might be related to growth, development and the reproduction regulation in sheep, which could provide a scientific basis for elucidating the genetic mechanisms of high reproduction in sheep.
