Project description:Metastasis accounts for 90% of cancer-related death. Distant metastasis follows a non-random pattern, termed “organotropism”, for instance, breast cancer (BC) preferentially metastasizes to the lung, bone, brain, and liver. The lung is a common site for BC metastasis, yet the underlying mechanisms and treatment strategies for lung metastasis remain unclear. To uncover the molecular mechanisms underlying the lung-tropic metastasis of breast tumors, we performed transcriptome sequencing on primary tumors and lung metastases.

Project description:Metastasis accounts for most of cancer-related deaths. Paracrine signaling between tumor cells and the stroma induces changes in the tumor microenvironment required for metastasis. Transcription factor c-Myb was associated with breast cancer (BC) progression but its role in metastasis remains unclear. Here we show that increased c-Myb expression in BC cells inhibits spontaneous lung metastasis through impaired tumor cell extravasation. On contrary, BC cells with increased lung metastatic capacity exhibited low c-Myb levels. We identified a specific inflammatory signature, including Ccl2 chemokine; that was expressed in lung metastatic cells but was suppressed in tumor cells with higher c-Myb levels. Tumor cell-derived Ccl2 expression facilitated lung metastasis and rescued trans-endothelial migration of c-Myb overexpressing cells. Clinical data show that the identified inflammatory signature, together with a MYB expression, predicts lung metastasis relapse in BC patients. These results demonstrate that the c-Myb-regulated transcriptional program in BCs results in a blunted inflammatory response and consequently suppresses lung metastasis.

Project description:Stephen Paget first proposed, in 1889, that organ distribution of metastases is a non-random event, yet metastatic organotropism remains one of the greatest mysteries in cancer biology. Here, we demonstrate that exosomes released by lung-, liver- and brain-tropic tumor cells fuse preferentially with resident cells at their predicted destination, such as fibroblasts and epithelial cells in the lung, Kupffer cells in the liver, and endothelial cells in the brain. We found that exosome homing to organ-specific cell types prepares the pre-metastatic niche and that treatment with exosomes derived from lung tropic models can redirect metastasis to the lung. Proteomic profiling of exosomes revealed distinct integrin expression patterns associated with each organ-specific metastasis. Whereas exosomal integrins α6β4 and α6β1 were associated with lung metastasis, exosomal integrins αvβ5 and αvβ3 were linked with liver and brain metastases, respectively. Targeting α6β4 and αvβ5 integrins decreased exosome uptake and metastasis in the lung and liver, respectively. Importantly, we demonstrate that exosome uptake activates a cell-specific subset of S100 family genes, known to support cell migration and niche formation. Finally, our clinical data indicate that integrin-expression profiles in circulating plasma exosomes from cancer patients could be used to predict organ-specific metastasis. Project description:Approximately 50% of patients with surgically resected early-stage lung cancer develop distant metastasis. At present, there is no in vivo metastasis model to investigate the biology of human lung cancer metastases. Using well-characterized patient-derived organoids (PDOs) from patients with lung adenocarcinoma (LUAD), we establish an in vivo metastasis model that preserves the biologic features of human LUAD metastases. Results of whole-genome and RNA sequencing performed in this study establish that our in vivo PDO metastasis model can be used to study clonality and tumor evolution and to identify biomarkers related to organotropism. Investigation of the response of KRASG12C PDOs to Sotorasib demonstrates that the model can examine the efficacy of treatments to suppress metastasis and identify mechanisms of drug resistance. Finally, our PDO model cocultured with autologous peripheral blood mononuclear cells can potentially be used to determine the optimal immune-priming strategy for individual patients with LUAD.

Project description:Clinical management of breast cancer (BC) metastasis remains an unmet need as it accounts for 90% of BC-associated mortality. Although the luminal subtype, which represents >70% of BC cases, is generally associated with a favorable outcome, it is susceptible to metastatic relapse as late as 15 years after treatment discontinuation. Seeking therapeutic approaches as well as screening tools to properly identify those patients with a higher risk of recurrence is therefore essential. Here, we report that the lipid-degrading enzyme fatty acid amide hydrolase (FAAH) is a predictor of long-term survival in patients with luminal BC, and that it blocks tumor progression and lung metastasis in cell and mouse models of BC. Together, our findings highlight the potential of FAAH as a biomarker with prognostic value in luminal BC and as a therapeutic target in metastatic disease.

Project description:While primary breast cancer (BC) is often effectively managed, metastasis remains the primary cause of BC-related fatalities. Gaps persist in our understanding of the mechanisms underlying cancer cell predilection for specific organs (organotropism). Unraveling these mediators of site-specific metastasis could enhance early detection and enable more tailored interventions. Liquid biopsy represents an innovative diagnostic approach in cancer involving the non-invasive analysis of biological materials found in body fluids like blood or urine. These materials include circulating tumor cells, cell-free circulating tumor DNA, and tumor-derived extracellular vesicles (EVs), which offer valuable insights for characterizing and monitoring tumor genomes to advance personalized medicine. EVs, lipid-bound particles released from cells into the extracellular space, facilitate intercellular communication by transporting proteins and DNA (EV-DNA) implicated in cancer progression and organ-specific metastasis. The aim of this study was to conduct an in-depth analysis of cell-free DNA (cfDNA) and EVs using a murine model of BC cell lines with distinct metastatic potentials and organotropisms (lung, bone, liver, brain). We characterized the secretome of 8 different metastatic cell lines originating from the same parental non metastatic and metastatic cell line, as well as normal mammary epithelial cells to identify unique biomarkers specific to metastatic sites. Using ultracentrifigcation, we isolated small EVs ranging from 78.9 to 101nm isolated from the conditioned media of the cell lines. We confirmed the presence of established EV markers, including CD81, TSG101 and syntenin. Transmission electron microscopy revealed interesting EV secretion patterns that differed according to cell line. We demonstrated the presence of EV-DNA and cfDNA in all the cell lines, with varying concentrations. We also found that a common BC mutation in TP53 can be specifically detected in both EV-DNA and cfDNA of cancer cell cultures using droplet digital PCR. The proteomic characterization of EVs through high throughput mass spectrometry revealed common EV signatures and proteins involved in cancer processes and organotropism unique to distinct site-specific cell lines. Specifically, we found enrichment of integrin receptors in metastatic cancer EVs compared to EVs secreted from normal epithelial cells and matched tumorigenic nonmetastatic cell lines. Gene ontology (GO) pathway analysis revealed that cancer derived EVs display a cell adhesion signature and are enriched with proteins involved in extracellular matrix interaction, focal adhesion, endocytosis, and PI3K-Akt signaling pathway. Interestingly, EVs containing heat shock proteins and DNA binding proteins are released by both cancer and normal cells. A total of 460 proteins were quantified with a high confidence level, including 70% of the TOP 200 exosome markers archived by Vesiclepedia. Taken together, we characterized the quantity and cargo of EVs in a unique model of BC organotropism, suggesting that EV-DNA and EVs were informative of normal and cancer states. This work could help to identify BC biomarkers associated with site-specific metastasis as well as new therapeutic targets.

Project description:Cancer extracellular vesicles (EVs) mainly exert pro-tumoral functions by changing the phenotypes of stromal cells to the benefit of tumor growth and metastasis. In particular, they shuttle to distant organs and seed pre-metastatic niches facilitating subsequent colonization by circulating tumor cells. The levels of tumor secreted EVs have been correlated to tumor aggressiveness, however, the link between EV secretion mechanisms, their capacity to form pre-metastatic niches and the actual formation of metastasis remains obscure. Here, we show that two GTPases RalA and RalB control, through the phospholipase PLD1, the homeostasis of multi-vesicular bodies and thereby tune the biogenesis and secretion of a subtype of (pro-metastatic) EVs. Mice experiments revealed that RalA and RalB promote lung metastasis of mammary carcinoma cells without affecting their invasive behaviors. Importantly, we show in vivo that EVs from RalA or RalB depleted cells have limited organotropic capacities and, as a consequence, are less efficient in priming lung metastasis. Furthermore, we show that such EVs lack the adhesion molecule MCAM/CD146, which is responsible for EVs organotropism. Finally, we show that RalA and RalB have increased expression in human breast cancer patients with lung metastasis. Altogether, our study identifies Ral GTPases as central molecules linking the mechanisms of EVs secretion, cargo loading to their capacity to disseminate and induce pre-metastatic niches.

Project description:Bone is the most frequent site of metastases from breast cancer (BC), although no biomarkers are yet available to predict the risk of skeletal dissemination. Here, we attempted to identify a gene signature correlated with bone metastasis (BM) onset in circulating tumor cells (CTCs) isolated from 40 metastatic BC patients, grouped according to their metastasis sites at the time of enrollment, namely “BM” (bone-only), “ES” (extra-skeletal) or “BM+ES” (bone and extra-skeletal). A 134-gene panel, derived from an extensive literature review, was validated on sub-clones of the MDA-MB-231 BC cell line with variable organotropism and then applied to CTC groups for targeted RNA sequencing. No correlation was found between CTC number and clinico-pathologic variables, whereas 31 differentially expressed genes emerged from “BM” and “ES” CTC comparison. The putative prognostic meaning of the “top-10” most deregulated genes was finally explored and pointed out by employing an external BC dataset.

Project description:We identified 967 proteins among 75 serum samples from patients with BC. Among these, 39 proteins were altered in serum samples at diagnosis, between progressors and non-progressors. Among these, 4 proteins were further altered when the progressors developed distant metastasis. In addition, within progressors, 20 proteins were altered in serum collected at diagnosis versus at the onset of metastasis. Pathway analysis showed that these proteins encoded pathways that describe metastasis, including epithelial mesenchymal transition and focal adhesion that are hallmarks of metastatic cascade.
