Expression data from HEK-293 cells treated with different CDK8/19 inhibitors

ABSTRACT: Hepatitis delta virus (HDV) enters hepatocytes via the receptor NTCP and requires CDK8/19 Mediator kinases to establish its infection by RNA Polymerase II. Here, we are interested in assessing the class effect of various CDK8/19 inhibitors and investigating the possible off-target effects caused by them.

Transcriptome profiles of Huh7.5.1-NTCP cells treated with HBV preS1 peptide

Project description:Chronic hepatitis B, C and D virus (HBV, HCV, HDV) infections are leading causes of liver disease and cancer worldwide. Although these viruses differ markedly in their life cycle and genomic organization, they exclusively infect hepatocytes. Recently, the sodium taurocholate cotransporting polypeptide (NTCP) was identified as the first functional receptor for HBV and HDV. Here, we report that NTCP also facilitates HCV entry into human hepatocytes, by augmenting the bile acid-mediated repression of IFN-stimulated genes (ISGs), including IFITM2 and IFITM3, to increase the susceptibility of cells to HCV entry. Furthermore, an HBV-derived preS1 peptide, known to bind NTCP and to inhibit bile acid uptake and HBV infection, inhibits HCV entry by enhancing the expression of ISGs. Our study highlights NTCP as a novel player linking bile acid metabolism to the interferon response in hepatocytes and establishes a role for NTCP in the entry process of multiple hepatotropic viruses, via distinct mechanisms. Collectively, these findings enhance our understanding of hepatitis virus-host interactions and suggest NTCP as an attractive antiviral target for HBV/HCV co-infection. Comparative analysis of gene expression upon IFNα stimulation in hepatocyte-like cells [IFN]

Project description:Patients chronically co-infected by Hepatitis B Virus (HBV) and Hepatitis D Virus (HDV) suffer from the most aggressive form of viral hepatitis leading to severe liver disease such as cirrhosis, hepatocellular carcinoma and liver decompensation. Treatments options are very limited with only two drugs used in clinic (IFN-α and Bulevertide) that rarely allow viral clearance. The molecular mechanisms leading to inhibition of HDV by IFN-α (in patients but also in vitro) are not known and mechanisms behind treatment failures in patients also remain elusive. Here we aimed at the identification of interferon stimulated genes (ISGs) that can specifically inhibit intracellular HDV replication. IFN-α was able to inhibit HDV in both co-infected differentiated HuH7.5-NTCP- and HepaRG cells (dHuH7.5-NTCP and dHepaRG, respectively). We hypothesized that ISGs responsible for the antiviral effect of HDV should be upregulated in both dHepaRG and dHuH7.5-NTCP cells upon treatment with IFN-α. To identify these ISGs, dHuh7.5-NTCP and dHepaRG cells were stimulated with IFN-α for 8h, total RNAs were extracted and gene expression was analyzed by RNAseq.

Confirmation of cdk8/19 inhibitor selectivity and activity in colorectal cancer

Project description:The Mediator complex is an evolutionarily conserved regulator of gene expression, influencing chromatin structure and RNA polymerase II-mediated transcription. Its activity is modulated by a protein kinase module, which includes cyclin-dependent kinases 8 and 19, that phosphorylate RNA polymerase II and transcription factors to regulate gene expression. CDK8 exhibits both oncogenic and tumour-suppressive activities depending on the cellular context, while the contributions of CDK19 remain less well understood. Based on prior data with our CDK8/19 inhibitor - 3,4,5-trisubstituted pyridine compound CCT251545 - we selected the human colorectal cancer cell line COLO205 for profiling experiments. This model is highly dependent on mutant BRAF and oncogenic β-catenin-driven WNT signaling and exhibits high basal β-catenin-dependent TCF/LEF promoter activity, which is sensitive to CDK8/19 inhibition in vitro and in vivo.
