Project description:Mechanical energy–driven portable water disinfection has attracted attention for its electricity-free operation, but this approach generally faces bottlenecks such as a high mechanical activation threshold, energy dispersion, and low interfacial reaction efficiency, making it difficult to achieve rapid and stable pathogen inactivation in practical scenarios. Here we report a manually operated portable water disinfection system that can inactivate 99.9999% of V. cholerae within 1 minute and demonstrate broad-spectrum disinfection against bacteria, fungi, parasites, and viruses. Amino-modified SiO₂ nanoparticles loaded with Au nanoparticles capture hydrated electrons and transfer them to the electret surface to generate localized nanoscale electric fields, which are further strengthened by hydrophobic fluorinated groups. This interfacial architecture not only promotes charge accumulation and transfer, but also leverages the intensified electric field to actively drive reactive oxygen species (ROS) generation at the solid–liquid–air interface, thereby markedly enhancing disinfection rate and efficacy compared with existing contact electrification–based disinfection technologies. Owing to its ease of operation, ourinterfacial electric-field-enhanced (IEFE) disinfection system is readily deployable in disaster relief and resource-constrained regions.

Project description:Goal was to identify yeast genes whose expression changed as a function of the shift from growth in bulk culture to growth in an air-liquid interfacial biofilm. Experiment Overall Design: Cells were grown 24 h at 30 C in YEPD to a density of about 500,000,000 cells/ml. At harvest, sugar was found to be depleted as measured by an enzymatic dip stick (Diastic, Bayer). Cells were pelleted and washed twice in sterile distilled water by centrifugation and then diluted 10-fold into 100 ml of Flor medium (YNB + 4% ethanol + leucine + histidine + uracil) in triplicate 250 ml beakers. Cultures were then grown statically in Flor medium at 27 C. Within a few hours following inoculation, the bulk liquid appeared to be clear, no biofilm was evident by visual observation, and a layer of settled cells was evident at the bottom of the beakers. After 48 h, a visible air-liquid interfacial biofilm covered the entire surface while the thickness of the layer of settled cells appeared unchanged. After 48 h, biofilm cells (floaters) were collected by aseptic aspiration. Once the biofilm cells were removed, cells at the bottom of the beaker (sinkers) were collected similarly. Cells from both populations were washed once in sterile distilled water by centrifugation prior to RNA isolation. Significant cell clumping was evident in both populations of cells by microscopic observation. While cell viability was estimated by plating on YEPD, the resultant cfu/ml values could not be directly correlated with cell counts in a hemacytometer because clumps of cells containing at least one viable cell presumably produced only a single colony. Further, counting individual cells accurately in the numerous clumps containing large numbers of cells was not possible. Nonetheless, when cells were counted (clumps were counted as single cells) and compared to cfu/ml for the same suspension, the cfu/ml values were about 2-fold higher than the corresponding values for cell counts using the hemacytometer for both biofilm and bottom layer cells.

Project description:During the past decades, there have been major advances in the field of biomaterials, thereby generating a vast variety of materials for a broad range of tissue engineering and regeneration applications. Although gene expression profiling has been used occasionally in biomaterial research, its usefulness for understanding cell-biomaterial interactions should be further explored for it to fulfill its promise as a tool to assess and improve material properties. Here, the transcriptional landscape induced by 23 materials is explored with a variety of properties within the scope of bone regeneration. An osteoblast cell line is used to identify the gene expression profiles that can be adopted in response to biophysical and chemical cues. It is shown that TGF-beta and WNT signaling may be involved in the cellular response to osteoinductive materials along with differential cell adhesion kinetics via attenuated FAK signaling. The previously reported effect of calcium and phosphate on BMP2 and TGF-beta signaling is confirmed and the biological effect of the addition of nanohydroxyapatite in poly (d,l-lactic acid) polymer particles is studied. Together with future applications, this approach will help researchers understand cellular responses in relation to material properties, which will promote the development of more effective biomaterials for applications in tissue regeneration.

Project description:Cell-cell adhesion is crucial for maintaining cell functions and the integrity of tissue structure in organisms. However, cell-cell adhesion cue has not been effectively replicated in biomaterials and cell-cell adhesion associated mechanisms that enhance neural regeneration remained largely unexplored. Here, we present a diffusive N-cadherin functionalized hydrogel system, which provided cell-cell adhesion cue to modulate intercellular communications to significantly promote the formation of active neural network. The dynamic assembly of N-cadherin at cell membrane-hydrogel interface driven by adhered cells occurred at a timescale of minutes, facilitating the reshaping of membrane protrusions to initiate intercellular adherens junctions and modulate long-term cell-cell communications. Focusing on the mechanisms, it is associated with Thrombospondin-1 (THBS-1) mediated neural communication and activation of TGF-β/Smad pathway. Further, this hydrogel system promisingly promoted neurological function recovery in rats following traumatic brain injury. Our study provided the principles of replicating diffusive cell adhesion molecules to mediate cell-cell adhesion in hydrogels, which may have broad applications in the development of engineered biomaterials aimed at modulating cell fates in regeneration of various tissues.

Project description:Scaffold vascularization plays a pivotal role in the wound healing process, facilitating the recruitment of endogenous progenitor cells and delivering crucial signaling molecules that promote cellular invasion, angiogenesis, and neo-tissue formation. In this study, we introduce nanosilicates as pro-angiogenic biomaterials that can prime endogenous cells to expedite angiogenesis and graft vascularization in vivo. Characterized by their mineral-based two-dimensional (2D) structure and extensive surface area, nanosilicates are efficiently internalized by endothelial cells, leading to augmented cellular migration and the formation of tubular structures. Through whole transcriptome sequencing, we elucidated that nanosilicates activate the canonical Wnt pathway through hypoxia-induced ROS production. In vivo investigations further corroborate the pro-angiogenic efficacy of nanosilicates-loaded biomaterials. This study posits nanosilicates as a potential pro-angiogenic adjunct in the design of biomaterials for in situ tissue regeneration.
