Project description:Attachment security is theorized to shape stress reactivity, but extant work has failed to find consistent links between attachment security to mothers and infant cortisol reactivity. We examined family configurations of infant-mother and infant-father attachment security in relation to infant cortisol reactivity. One-year old infants (N = 180) participated in the Strange Situation with mothers and fathers in two counterbalanced lab visits, one month apart (12 and 13 months). Infants with secure attachments only to their fathers and not their mothers had higher cortisol levels than infants with a secure attachment to mother and also exhibited a blunted cortisol response (high at baseline and then a decrease after stress). Results suggest that a secure attachment to father may not be enough to reduce infant stress reactivity when the infant-mother attachment is insecure, and future research is needed to uncover the family dynamics that underlie different family configurations of attachment security.

Project description:It is well established that faces evoke a distinct neural response in the adult and infant brain. Past research has focused on how the infant face-sensitive ERP components (N290, P400, Nc) reflect different aspects of face processing, however there is still a lack of understanding of how these components reflect face familiarity and how they change over time. Further, there are only a few studies on whether these neural responses correlate with other aspects of development, such as infant temperament. In this longitudinal study (N∼60), we recorded infant visual ERPs in response to mother and stranger face stimuli at 4, 6 and 9 months of age. Our results showed that, compared to a stranger face, the mother face evoked a larger N290 at 4 months and a larger P400 at 6 months. At 9 months, no difference was found between mother and stranger faces. However, at 9 months we found that the P400 and Nc amplitudes evoked by the mother face were associated with infant falling reactivity. We conclude that the neural responses associated with the processing of faces, and specifically the face of the mother, are related to the development of infant individual characteristics.

Project description:We used the Virochip microarray to detect and discover a novel adenovirus, TMAdV (titi monkey adenovirus). We used custom-commercial microarrays from Agilent Technologies. The microarray platform GPL11662 consists of 62,976 probes, of which 19,058 probes are 70-mer viral oligonucleotides (the union of the V3 and V4 platforms -- please see Wang, et al., PNAS, 2002; Chiu, et al., Clin Infect Dis, 2006; Chiu, et al., PNAS, 2008). For this study, 4 experimental Virochip microarrays derived from tissue and swabs from affected titi monkeys were analyzed. We also used 18 Virochip microarrays derived from sera from healthy random human blood donors as a negative background set for Z-score analysis (see Chiu, et al., Clin Infect Dis, 2006). Raw Agilent microarray .txt files are background normalized and the 19,058 70-mer viral probes are then selected and displayed as columns in the processed data table.

Project description:Past research has focused on infants' visual preference for the mother's face, however it is still unknown how these responses change over time and what factors associate with such changes. A longitudinal study (N ~ 60) was conducted to investigate the trajectories of infant visual preference for the mother's face and how these are related to the development of emotional reactivity in the first year of life. Two face stimuli (i.e., the infant's mother and a consistent stranger face) were used in a visual preference task at 2 weeks, 4, 6, and 9 months of age. At each time point, mothers were asked to complete a measure of infant temperament via standardised questionnaires. Our results show that while at 2 weeks, 4 months and 9 months of age infants looked equally at both faces, infants at 6 months looked significantly longer at their mother's face. We also observed prospective associations with emotional reactivity variables so that infants who looked longer at the mother's face at 6 months showed higher falling reactivity, i.e. a better ability to recover from distress, at 9 months. We discuss these findings in light of the roles that both infant development and the caregiver play in emerging emotion regulation capacities during the first year of life.

Project description:ObjectiveA number of studies have established an association between antenatal maternal depression and infant sleep. One key question is whether all infants are equally susceptible to environmental influences, including the intrauterine environment. Reactive temperament has been examined as a plasticity factor, with accumulating evidence suggesting that infants with reactive temperament may be more susceptible to both positive and negative environmental influences. This study examines whether infant reactivity moderates any association between antenatal depression and infant sleep in 2 longitudinal studies: the Avon Longitudinal Study of Parents and Children (ALSPAC) and Generation R cohorts.MethodsMaternal depression scores were assessed during pregnancy using Edinburgh Postnatal Depression Scale and Brief Symptom Inventory. Infant sleep duration and awakenings, in ALSPAC (N = 8318) and Generation R (N = 2241), were assessed at 18 and 24 months of age, respectively. Infant reactivity was assessed by temperament questionnaire at 6 months of age.ResultsHierarchical linear regression models indicated a 3-way interaction between reactivity and gender moderating the effect of antenatal depression on infant sleep, on sleep duration in Generation R at 24 months (β = .085, p < .001) in the whole sample and when limited to the Dutch/European group (β = .055, p = .030), and on night awakenings at 18 months in ALSPAC (β = -.085, p = .013). Boys with more reactive temperament exhibited shorter sleep duration and a higher number of awakenings when previously exposed to maternal symptoms of antenatal depression.ConclusionFor the first time, these findings highlight, in 2 large cohorts, that children with temperamental reactivity may be more vulnerable to antenatal depression, raising the possibility of targeted interventions to improve infant outcomes.

Project description:Maternal biological systems impact infant temperament as early as the prenatal period, though the mechanisms of this association are unknown. Using a prospective, longitudinal design, we found that maternal (N = 89) amplitudes of the late positive potential (LPP) in response to negative stimuli during the second, but not the third, trimester of pregnancy predicted observed and physiological indices of temperamental reactivity in infants at age 4 months. Maternal LPP was positively associated with observed infant fear and negatively associated with frontal EEG asymmetry and cortisol reactivity in infants at age 4 months. Results identify a putative mechanism, early in pregnancy, for the intergenerational transmission of emotional reactivity from mother to infant.

Project description:Significance: Cerebrovascular reactivity (CVR), defined as the ability of the cerebral vasculature to dilate or constrict in response to a vasoactive stimulus, is an important indicator of the brain's vascular health. However, mechanisms of cerebrovascular dysregulation are poorly understood, and no effective treatment strategies for impaired CVR exist. Preclinical murine models provide an excellent platform for interrogating mechanisms underlying CVR dysregulation and determining novel therapeutics that restore impaired CVR. However, quantification of CVR in mice is challenging. Aim: We present means of assessing CVR in awake mice using intraperitoneal injection of acetazolamide (ACZ) combined with continuous monitoring of cerebral blood flow. Approach: Measurements of cerebral blood flow were made with a minimally invasive diffuse correlation spectroscopy sensor that was secured to an optical window glued to the intact skull. Two source-detector separations (3 and 4.5 mm) per hemisphere were used to probe different depths. CVR was quantified as the relative increase in blood flow due to ACZ. CVR was assessed once daily for 5 days in 5 mice. Results: We found that CVR and the response half-time were remarkably similar across hemispheres and across 3- versus 4.5-mm separations, suggesting a homogenous, whole brain response to ACZ. Mean(std) intra- and intermouse coefficients of variations were 15(9)% and 19(10)%, respectively, for global CVR and 24(15)% and 27(11)%, respectively, for global response half-time. Conclusion: In sum, we report a repeatable method of measuring CVR in free-behaving mice which can be used to screen for impairments with disease and to track changes in CVR with therapeutic interventions.
