Gene expression data of distal colon mucosa from young and old human

ABSTRACT: The disruption of the mucosal barrier of the digestive tract is a common pathological change in the elderly, which often causes inflammatory bowel disease among old people. Given that RNA molecules are dysregulated in numerous diseases, we performed high‑throughput analysis to compare RNA expression profiles in normal distal colon mucosa between young and aged individuals. Samples were obtained from 3 young adults (27, 28, and 36 years of age) and 3 aged adults (72, 79, and 81 years of age).

A novel model of binge ethanol exposure reveals enhanced neurodegeneration with advanced age

Project description:Binge drinking is rising among aged adults (>65 years of age). The distinct effects of binge ethanol exposure on neurodegeneration in the aged brain are not well described. Using our model of intermittent binge ethanol exposure in young and aged mice, we investigated how binge ethanol expoure may promote neurodegenerative diease development. Spatial transcriptomics revealed that binge ethanol exposure enriched neurodegenerative disease pathways in the aged hippocampus, including tau hyperphosphorylation and neuronal death. These data suggest binge drinking during advanced aging may promote neurodegenerative disease development.

Project description:This study examines how aging influences the proteomic landscape of human spermatozoa. Proteomic analysis was done on sperm samples from three groups of normal, fertile men: young adults (21–30 years), late adults (31–40 years), and advanced age (41–51 years). The analysis revealed dynamic age-related alterations in human sperm proteins potentially impacting key sperm physiological functions such as motility, metabolism, and fertilization.

Tear proteomics of young patients wearing glasses, orthokeratology lenses, and soft contact lenses

Project description:Contact lens-related ocular surface complications occur more often in teenagers and young adults. The purpose of this study was to determine changes in tear proteome of young patients wearing glasses (GL), orthokeratology lenses (OK), and soft contact lenses (SCL). Twenty-two young patients (10-26 years of age) who were established (> 3 years) GL (n=10), OK (n=6), and SCL (n=6) wearers were recruited. Tears were collected via Schirmer strips. Proteomic data were collected using a data-independent acquisition-parallel accumulation serial fragmentation (diaPASEF) workflow. Tear proteome composition and abundance were compared across different correction groups and between the children (age <18 years) and young adults (age 18 years) GL wearers. We identified 3406 protein groups in tears. Among proteins identified in 80% of tear samples, 8 proteins were upregulated, and 11 proteins were down-regulated in the SCL group compared to the OK group. Eighty-two proteins were differentially expressed in children and young adults GL wearers, among which 67 proteins were upregulated, and 15 proteins were downregulated in children. These 82 proteins were involved in 1 inflammation, 9 immune, and 1 glycoprotein metabolic biological processes. As teenagers and young adults have the highest risk of developing contact lens-related complications, this work highlights the importance of understanding ocular responses to contact lens wear across different ages.

Project description:The objective was to determine age-associated changes in the transcriptome of granulosa cells recovered from the dominant follicle at the time of selection. Granulosa cells were collected from the dominant follicle of aged and young cows after ovariectomy (15 Â±1.5 years, n=3 and 6 Â± 1.1 years, n=3), or ultrasound-guided follicle aspiration (16Â±2.1 years, n=4 and 7Â±1.2 years, 6 Â± 1.1 years, n=4) 3-days after ovulation (Day 0). Messenger-RNA was extracted, amplified, labeled with florescent dyes, and hybridized with bovine-specific microarrays (GEO accession # GPL13226). Target intensities were analyzed to determine differential gene expression in granulosa cells from aged vs. young cows. A total 169 genes were differentially expressed (â¥ 2 fold-change; Pâ¤0.05) between age groups. In conclusion, granulosa cells collected at the time of selection of the dominant follicle exhibited age-related changes in the transcriptome that may explain follicle-associated loss of oocyte competence in aged cows. Gene Expression Data in Human Monocyte-derived Dendritic Cell (MoDC) from Young and Aged Donors

Project description:Dendritic cells (DCs) are major antigen-presenting cells that play a key role in initiating and regulating innate and adaptive immune responses. DCs are critical mediators of tolerance and immunity. The functional properties of DCs changes with age. The purpose of this study was to define the age-associated molecular changes in DCs by gene array analysis using Affymatrix GeneChips. We identified up and down-regulated gene expression changes in DC from aged donors compared to young donors. Total 9 MoDC RNA samples from four young donors (20-27 years) and five aged donors (77-84 years) were analyzed using Affymetrix HG-U133A_2 Gene Arrays to compare differential gene expression changes in MoDC between aged and young groups
