The effects of oleic acid diet on ovarian cancer tumor growth in murine xenograft model

ABSTRACT: Oleic acid is an unsatuarated fatty acid that is converted by SCD1 (Stearoyl-CoA Desaturase 1), which is highly expressed in in ovarian cancer and has protective effect on cancer cell survival. We here report oleic acid high diet has upregulated serveral cell cycle related pathways such as E2F targets, mitotic spindle and G2M targets to promote cell proliferation in murine xenograft model.

Oleic Acid Availability Preprograms Early Thymocytes for Treg Cell Commitment in the Periphery

Project description:The natures of activation signals are essential in determining T cell subset differentiation, however, the properties of T cell subset preference acquired during intrathymic development remain elusive. Here we show that naïve CD4+ T cells generated in the thymic microenvironment lacking SCD1, the enzyme catalyzing oleic acid production, exhibit enhanced Treg cell differentiation and confer on mice the ability to resist EAE. SCD1 deletion in K14+ thymic epithelia recapitulated the enhanced Treg cell differentiation phenotype of SCD1 knockout mice. The dearth of oleic acid permitted Dot1L to increase H3K79me2 level at the Atp2a2 locus of thymocytes at the DN2-DN3 transition stage. Such epigenetic modification persisted to naïve CD4+ T cells and facilitated Atp2a2 expression. Upon TCR activation, Atp2a2 enhanced the activity of calcium-NFAT1-Foxp3 axis to promote naïve CD4+ T cells to differentiate into Treg cells. Therefore, oleic acid availability is critical for preprogramming thymocytes with Treg differentiation propensities in periphery.

SCD1 Modulates the Skin Homeostasis via Sustaining Hemidesmosomes

Project description:Niche for stem cells can profoundly influence the position and fate of stem cells in tissue homeostasis and associated disorders, however, the mechanisms underlying the maintenance of stem cell niche remain poorly understood. Here we report that fatty acid desaturation catabolized by stearoyl-coenzyme A desaturase 1 (SCD1) regulates the activity of hair follicle stem cells (HFSCs) via blocking the formation of the bulge, a niche for HFSCs. We further unraveled that the abnormal hair growth in mice with SCD1 deletion is due to its function in K14+ basal keratinocytes rather than directly affecting HFSCs. Mechanistically, SCD1 deficiency impaired the expression of integrin α6 and the formation of hemidesmosomes (HDs), and thus allowed the activation of FAK in K14+ keratinocytes. Activated FAK subsequently stimulated PI3K and promoted the differentiation and proliferation of these keratinocytes, resulting in downward extension of the outer root sheath (ORS) and the disruption of bulge formation. Interestingly, inhibition of PI3K signaling restored bulge formation and normalized HFSCs and hair growth. Supplementation of oleic acid, the product of SCD1, to SCD1-/- mice also enhanced HD numbers, normalized the niche of HFSCs, and promoted hair growth in SCD1-/- mice. Taken together, SCD1 is indispensable for hair growth through stabilizing the number of HDs and thus sustaining bulge formation for HFSC residence and periodic activity.

Regulation of stress signaling by SCD1

Project description:Cytotoxic stress activates stress-activated kinases, initiates adaptive mechanisms, including the unfolded protein response (UPR) and autophagy, and induces programmed cell death. Fatty acid unsaturation, controlled by stearoyl-CoA desaturase (SCD)1, prevents cytotoxic stress but the mechanisms are diffuse. We found that SCD1 inhibition alters the levels of proteins phosphorylated at tyrosine in fibroblasts. Major regulated proteins were identified following immunoprecipitation using an anti-phospho-tyrosine antibody and subsequent proteomic analysis.

Project description:Compared to ordinary rapeseed, high-oleic acid rapeseed has higher levels of monounsaturated fatty acids and lower levels of saturated fatty acid and polyunsaturated fatty acids, and thus is of high nutritional and health value. In addition, high-oleic acid rapeseed oil imparts cardiovascular protective effects. Based on these properties, high-oleic acid oil crops have been extensively investigated and cultivated. In this study, we employed a microarray analysis with high oleic acid line and low oleic acid line from the developing seeds (27 days after flowering) of Brassica napus.

Transcription profiling of livers of stearoyl-CoA desaturase 1-deficient mice to search for gene expression changes unique to SCD1-/- mice on a very low-fat, high- carbohydrate diet.

Project description:Stearoyl-CoA desaturase 1-deficient (SCD1-/-) mice have impaired monounsaturated fatty acid (MUFA) synthesis. When maintained on a very low-fat, high-carbohydrate (VLF-HC) diet, SCD1-/- mice develop severe hypercholesterolemia characterized by an increase in apolipoprotein B-containing lipoproteins and the appearance of lipoprotein-X. Additionally, high-density lipoprotein cholesterol is dramatically reduced in VLF-HC SCD1-/- mice. The concomitant presence of elevated plasma bile acids, bilirubin and aminotransferases in the VLF-HC SCD1-/- mouse are indicative of hepatic dysfunction. Supplementation of the VLF-HC diet with unsaturated fat (canola oil), but not saturated fat (coconut oil), prevents these plasma phenotypes. However, dietary oleate was not as effective as canola oil in reducing low-density lipoprotein cholesterol, signifying an additional role for dietary polyunsaturated fatty acid deficiency in the development of this phenotype. These results indicate that lack of SCD1 results in an increased requirement for dietary unsaturated fat to compensate for impaired MUFA synthesis and to prevent hypercholesterolemia and hepatic dysfunction. Experiment Overall Design: We used Affymetrix Mouse430v2.0 microarray chips to search for gene expression changes unique to the VLF-HC SCD1-/- group. These chips contain 45,101 probe sets representing over 39,000 transcripts and variants from over 34,000 mouse genes. We studied SCD1+/+ and SCD1-/- mice on chow or the VLF-HC diet, analyzing five individual livers from each subgroup for a total of twenty microarrays.

Project description:Bone marrow mesenchymal stem cells (BM-MSCs) are of multi-differentiating potential and has been demonstrated to have the ability to differentiate into endothelial cells. Stearoyl-CoA desaturase 1 (SCD1) is a key enzyme in lipid metabolism, which can convert saturated fatty acids into unsaturated fatty acids. We used lentivirus to transfected SCD1 gene into BM-MSCs and found that overexpression of SCD1 could promote the differentiation of BM-MSCs into endothelial cells. We used microarrays to detect the differential gene expression profile of endothelial induced BM-MSCs with SCD1 overexpression.

Department of Biochemistry, University of Wisconsin, Madison, WI 53706, USA. ntambi@biochem.wisc.edu Stearoyl-CoA desaturase (SCD) is a central lipogenic enzyme catalyzing the synthesis of monounsaturated fatty acids, mainly oleate (C18:1) and palmitoleate (C16:1), which are components of membrane phospholipids, triglycerides, wax esters, and cholesterol esters. Several SCD isoforms (SCD1-3) exist in the mouse. Here we show that mice with a targeted disruption of the SCD1 isoform have reduced body adiposity, increased insulin sensitivity, and are resistant to diet-induced weight gain. The protection from obesity involves increased energy expenditure and increased oxygen consumption. Compared with the wild-type mice the SCD1-/- mice have increased levels of plasma ketone bodies but reduced levels of plasma insulin and leptin. In the SCD1-/- mice, the expression of several genes of lipid oxidation are up-regulated, whereas lipid synthesis genes are down-regulated. These observations suggest that a consequence of SCD1 deficiency is an activation of lipid oxidation in addition to reduced triglyceride synthesis and storage. Keywords: Genetic modifications
