Activation of the JNK–ENO1 Axis Promotes Glycolytic Reprogramming and Progression of Retroperitoneal Leiomyosarcoma


ABSTRACT: Retroperitoneal leiomyosarcoma (RPLMS) is a highly aggressive malignancy with limited therapeutic options, and its metabolic underpinnings remain poorly understood. In this study, we identified significant activation of glycolytic pathways in RPLMS through RNA-seq analysis, with enolase 1 (ENO1) markedly upregulated in both tumor tissues and patient-derived cell lines. Functional experiments demonstrated that ENO1 knockdown suppresses proliferation, migration, and glycolytic metabolism in RPLMS cells. Mechanistically, JNK signaling was shown to directly promote ENO1 transcription, thereby driving glycolytic reprogramming. Our findings reveal that the JNK–ENO1 axis plays a critical role in RPLMS progression, suggesting that targeting this pathway may represent a promising therapeutic strategy for this devastating disease.
