Pleuromutilin Antibiotics Suppress Hepatocellular Carcinoma Growth

ABSTRACT: Pleuromutilin antibiotics(Lefamulin, Tiamulin, et al) has application potential in malignancies treatment. To mechanistically investigate the inhibitory activity against HCC of Lefamulin and Tiamulin, we treated Hepa1-6 cells with 37 μg/ml Lefamulin or Tiamulin for 2 days. Then the total RNA was performed RNA-Seq analysis.

Lefamulin targets ILF3 to overcome targeted therapy resistance in hepatocellular carcinoma

Project description:Acquired resistance represents a critical clinical challenge to molecular targeted therapies such as tyrosine kinase inhibitors (TKIs) treatment in hepatocellular carcinoma (HCC). Therefore, it is urgent to explore new mechanisms and therapeutics that can overcome or delay resistance. Here, we identified a U.S. Food and Drug Administration (FDA)-approved pleuromutilin antibiotic that overcomes sorafenib resistance in HCC cell lines, cell line-derived xenograft (CDX) and hydrodynamic injection mouse models. We demonstrated that lefamulin targets and binds Interleukin Enhancer-binding Factor 3 (ILF3) to inhibit sorafenib-induced mitochondrial ribosomal protein L12 (MRPL12) upregulation. Mechanistically, lefamulin directly binds to Ala-99 of ILF3 and interferes with acetyltransferase general control non-depressible 5 (GCN5) and CREB binding protein (CBP) mediated acetylation of Lys-100, which disrupting the ILF3-mediated transcription of MRPL12 and subsequent mitochondrial biogenesis. Clinical data further confirmed that high ILF3 or MRPL12 expression was associated with poor survival and targeted therapy efficacy in HCC. Conclusively, our findings suggest that ILF3 is a potential therapeutic target for overcoming resistance to TKIs, and lefamulin may be a novel combination therapy strategy for HCC treatment with sorafenib and regorafenib.

GPR35+ tumor-associated neutrophils promote tumor growth in hepatocellular carcinoma.

Project description:Bulk RNA-seq profiling of tumor-associated neutrophils from Hepa1-6 mouse hepatocellular carcinoma (HCC) models. Neutrophils were isolated from mice HCC tissues and classified as GPR35+ or GPR35- subsets (n=3 per group). Differential expression analysis revealed significant upregulation of MAPK signaling pathway and pathways associated with neutrophil migration and cytokines production in GPR35+ neutrophils. These data support the pro-tumorigenic role of GPR35+ neutrophils in HCC.

Antibiotics treatment ameliorates TET2 loss-of-function associated hematological malignancies

Project description:TET2 is one of the most frequently mutated genes in hematological malignancies. TET2 mutations are also frequently observed in healthy individuals with clonal hematopoiesis. Additional factors, such as inflammatory stress, might promote the expansion and initiate the pre-leukemic condition of Tet2 deficient hematopoietic stem cells. Antibiotics treatment is frequently used in normal individuals and patients with hematological malignancies treatment to suppress infection-induced inflammation. However, prolonged antibiotics treatment resulted in bone marrow suppression and gut microbiota alteration. In our study, we observed that the expansion of Tet2 deficient myeloid cells are positively correlated with serum cytokine levels at pre-malignant stages. We then evaluated the effect of antibiotic treatment in Tet2 deficient myeloid and lymphoid tumors in vivo. We found that antibiotics treatment suppressed the growth of Tet2 deficient malignant cells in vivo. RNA-seq analysis revealed significant changes in immune related signaling pathways (e.g., Tnf-α signaling) in antibiotics treated Tet2 deficient myeloid and lymphoid tumor cells. Suppression of Tnf-α signaling using pharmacological inhibitors partially suppressed Tet2 deficient tumor cell growth in vivo. In summary, our results suggest that the expansion of Tet2 deficient blood cells are positively associated with a pre-inflammatory condition and suppression of inflammatory pathways may attenuate the progression of TET2 inactivation-associated hematological malignancies.

Project description:We wanted to measure the transcriptomic changes of P.aeruginosa (PAO1 strain) when treated with plant extracts (Melastoma dodecandrum Lour, and Potentilla indica) and antibiotics (triclosan, rifampicin, gentamicin and ciprofloxacin) against DMSO control to aid in understanding the mode of action of our plant extracts. PAO1 samples were treated with plant extracts and antibiotics at MIC, for 24h at 37°C with OD readings taken at 600 nm every 10 mins using a microplate reader.

XPO1inhibition induces NCOR1-Dependent oxidative stress to suppress hepatocellular carcinoma

Project description:Hepatocellular carcinoma (HCC) remains a therapeutic challenge due to the limited efficacy of current systemic therapies. To identify potential novel therapeutic approaches, computational analysis of HCC datasets and drug screening were integrated, leading to the repurposing of hematological cancer drug selinexor (an XPO1 inhibitor) for HCC treatment. Functional studies revealed that XPO1 inhibition triggers oxidative stress and cell cycle arrest in HCC cells through nuclear sequestration of NCOR1, disrupting redox homeostasis through FOXK1-dependent transcriptional activation of genes associated with Reactive Oxygen Species (ROS). A genome-wide CRISPR-Cas9 screen further identified the KEAP1-NRF2 axis as a key determinant of sensitivity to XPO1 inhibition. Furthermore, high-throughput compound screening demonstrated that disulfiram, a clinically-used aldehyde dehydrogenase inhibitor, synergizes with XPO1 inhibitor through exacerbation of ROS accumulation. Collectively, these findings demonstrate the therapeutic repurposing of selinexor for HCC while uncovering its novel mechanism of action, establishing a predictive biomarker, and proposing an immediately translatable combination therapy.

Transcriptome of hepatocellular carcinoma using CAGE

Project description:An increasing number of non-coding RNAs (ncRNAs) are implicated in various human diseases including cancer; however ncRNA transcriptome of hepatocellular carcinoma (HCC) remains largely unexplored. We use CAGE (Cap Analysis of Gene Expression) to comprehensively map transcription start sites (TSSs) across different etiologies of human HCC as well as mouse HCC, with particular emphasis on ncRNAs distant from protein-coding genes. We find thousands of significantly up-regulated distal ncRNAs in HCC tumors compared to their matched non-tumors, which are as many as protein-coding genes. Moreover, we identify many LTR retroviral promoters activated in HCC tissues and expressed in a subfamily-specific manner, which account for approximately 20% of the up-regulated distal ncRNAs. The transcripts derived from LTRs, determined by 3' RACE, are multi-exon nuclear ncRNAs typically 0.5-2kb in length. This study sheds light on ncRNA transcriptome of human and mouse HCC. Expression profiles using CAGE for 37 mouse HCC. The human data are archived at dbGaP (phs000885.v1.p1). In this study, our results demonstrated that the absence of Ezh2 in bone marrow-derived macrophages (BMDMs) exacerbated M2-like phenotypic changes upon incubation with conditioned media (CM) from hepatocellular carcinoma (HCC) cell line Hepa1-6. The ATAC-seq revealed that the promoter regions of pyruvate metabolic process-related genes were more accessible in treated BMDMs Ezh2 KO.
