Human Achilles tendon spatial transcriptomics

ABSTRACT: We used 10x Genomics Xenium spatial profiling to map gene expression across sections of human fetal Achilles tendons. By combining genetic data with spatial coordinates and histology images, we identified specific cell types—including tendon stem cells, various mature tendon cells, and cartilage cells—and characterized different types of immune cells (macrophages) within the tissue structure.

Effects of running for 1h on gene expression in Achilles tendon cells of SD rats

Project description:The Achilles tendon is the thickest tendon in the human body, and Achilles tendinopathy is its most prevalent disorder, often considered a consequence of overuse. While disruption to the collagen fibers represents a significant manifestation of Achilles tendinopathy, strikingly little is known about the mechanisms by which healthy tendon accumulates damage in vivo.As existing studies of tendon biomechanics and mechanobiology predominantly relied on in vitro or ex vivo experiments on isolated tissues, it is still largely unknown whether and how disruptions occur to the collagen molecules in healthy Achilles tendons following physiological activities. We reported the first RNA-seq analysis reflecting transcriptome changes in healthy rat Achilles tendons following running, providing a resource for future investigations in tendon mechanobiology and sports medicine.

Spatial transcriptomics of the achilles tendon harvested from the injury site with and without TrkA neuronal inhibition

Project description:Peripheral afferent neurons terminate at the surfaces of tendons, yet their role after injury beyond nociceptive functions remain unclear. Using transgenic animal models, sensory neurons were found to sprout after Achilles tendon injury in domains of Nerve growth factor (NGF) expression. Conditional deletion of Ngf in either myeloid or mesenchymal cell types led to tendon repair defects – findings phenocopied by inactivation of TrkA (Tropomyosin receptor kinase A) using a knockin mouse model. We sought to identify the signals modulated with neural TrkA inhibition. A combination of single cell and spatial transcriptomic analysis was performed on the Achilles injury site of TrkA^F592A animals and compared them to TrkA^WT.

Project description:Tendon integrity depends on controlled extracellular matrix production and adaptation to mechanical load. Dysregulated hypoxia-inducible factor 1α (HIF1α) signaling has been linked to human tendon disease. To investigate the effects of chronic HIF1α activation and to compare responses between load-bearing Achilles tendons and positional tail tendon fascicles, we generated a tendon-specific VHL knockout in SCX-positive cells and performed label-free proteomics to characterize matrix remodeling.

PI3K-Akt signaling regulates Scx-lineage tenocytes and Tppp3-lineage tendon sheath synovial cells in physiological tendon regeneration

Project description:We identify a PI3K-Akt signaling pathway which is specifically upregulated in neonatal murine injured Achilles tendons. PI3K-Akt signaling inhibition in neonatal murine Achilles tendon rupture model decreases cell proliferation and cell migration to the regenerating tendons in both Scx-lineage intrinsic tenocytes and Tppp3-lineage extrinsic tendon sheath synovial cells. Moreover, PI3K-Akt signaling inhibition decreases stemness and promotes mature tenogenic differentiation in both Scx-lineage and Tppp3-lineage cells. Collectively, PI3K-Akt signaling plays an important role in physiological tendon regeneration.

Structure-function Relationships in Human Tendons: Interfascicular Matrix Specialisation in the Achilles Tendon C4PR_LIV

Project description:Tendon is a highly aligned connective tissue, in which the macro-structure consists of collagen-rich fascicles surrounded by interfascicular matrix (IFM). In a series of recent studies in equine tissue, we have demonstrated specialisation of tendon composition, structure and mechanics to achieve the tendon’s functional requirements, specifically reporting extensive specialisation of the IFM region in the energy storing superficial digital flexor tendon. We have also demonstrated loss of functional specialisms with ageing, leading to a hypothesised new paradigm for tendinopathy, focused on the importance of the IFM. However, to date, there have been no studies focused on structure-function specialisation or the IFM in functionally distinct human tendons. Here, we compare the positional anterior tibialis tendon and energy storing Achilles tendon, performing a detailed analysis of the composition and mechanical properties of both fascicle and IFM regions, to test the hypothesis that the IFM in the energy storing Achilles tendon has specialised composition and mechanical properties, and that these specialisations are lost with ageing. We demonstrate that the IFM is specialised in the energy storing Achilles tendon, with greater elasticity and fatigue resistance than in the positional anterior tibialis tendon. While there were few age-related alterations in mechanics, we did identify age-related alterations in the IFM proteome of the Achilles tendon specifically, which is predicted to be regulated by TGF-beta signalling and may be responsible for the trend towards decreased fatigue resistance observed in the Achilles IFM with ageing.

Achilles tendon exhibits distinct transcriptomic changes with increased mitochondrial energy production under lunar gravity

Project description:Gravity plays a fundamental role in maintaining the structure and function of biological tissues, particularly within the musculoskeletal system. In space, the absence or reduction of gravitational forces leads to musculoskeletal deterioration, posing significant challenges for astronauts and individuals experiencing prolonged immobilization. Consequently, the effects of microgravity on muscle and bone have been extensively studied. Recent research highlights the crucial role of tendons in force transmission and mechanical stability; however, their transcriptomic response to reduced gravity remains largely unexplored. Here, we examined the adaptations of the Achilles tendons to lunar gravity (1/6 of Earth gravity) using RNA sequencing of tissue samples obtained from mice reared in the International Space Station for 25-26 days. Compared to quadriceps femoris muscles and brown adipose tissue, a greater number of genes were upregulated in the Achilles tendons under reduced gravity. Enrichment analyses revealed a significant upregulation of mitochondrial energy production in the Achilles tendons exposed to reduced gravity. Meanwhile, genes associated with extracellular matrix organization and collagen integrity were downregulated. Although histological examination showed no obvious structural differences between the lunar and Earth gravity groups, the expression of Atp5b, a subunit of ATP synthase, was enhanced in the lunar gravity group. Our findings suggest that mechanical loading regulates tendons by influencing mitochondrial energy production and extracellular matrix composition. These insights may contribute to therapeutic strategies for tendon degeneration associated with aging and disuse-related conditions.

Project description:We have compared the gene expression profile of rat Achilles tendon-derived stem cells in post-natal tendon development. Rat tendon stem/progenitor cells (TSPCs) were isolated at different stages of post-natal development: TSPCs-1d, TSPCs-7d and TSPCs-56d. TSPCs at different post-natal development stages (1d, 7d and 56d) were isolated, cultured and used for microarray analyses at passage 2. All TSPCs in this study were of isolated from more than one individual. Total RNA was extracted and fragmented biotin-tagged cRNA was hybridized to Rat Genome 230 2.0 Array.
