The purpose of the experiment is to study DNA repair after base editing in preimplantation human embryos. Key findings are a lack of chromosomal aneuploidies in base editing samples, contrasting with the outcomes after Cas9 cleavage at the same sites. Off target base editing is highly gRNA dependent. Base editing is efficient, and lacks the genotoxicity of Cas9.

Project description:Donated 2PN human zygotes were injected with a Cas9 RNP targeting pericentromeric and mid-arm sites on chromosomes 16 and X. The purpose of this experiments was to determine if CRISPR induced genetic changes (e.g. indels or chromosomal changes) differ based on the targeted chromosome and position in the chromosome (pericentromeric vs chromosome mid-arm targets). We found whole and segmental chromosome loss in over one third of chromosomes targeted by CRISPR/Cas9. Chromosome loss was seen after targeting different chromosomes, and different positions in the chromosome. These results highlight additional limitations of CRISPR/Cas9 genome editing, as well as potential mechanisms of origin of aneuploidies in the human embryo.

Project description:After the library was qualified, the library was pooled according to the effective concentration and the demand of target offline data, and was sequenced by Illumina platform. Since PABPN1L can bind the poly(A) tail, we propose that it participates in the process of post-transcriptional regulation and degradation of maternal mRNA. We investigated the mRNA changes in GV oocytes, MII oocytes, and fertilized eggs using RNA-seq. GV oocytes, MII oocytes and zygotes were collected and mixed from three mice of each genotypes separately.

Each group contained a total of 30 oocytes or zygotes. In the initial step, we spiked 2 × 106 mRNA-EGFP, transcribed in vitro, into each group.Then, RNA was extracted and detected.Reverse transcription was performed on the samples that met the effective concentration.After quality inspection, build the library.GV oocytes, MII oocytes and zygotes were collected from three different PABPN1L KO or WT mice seperately.

Zygotes segregate entire parental genomes in distinct blastomere lineages causing cleavage stage chimaerism and mixolpoidy

Project description:Chimaerism and mixoploidy define the presence of cell lineages with different parental genomes or different ploidy states in a single individual. Our knowledge on their mechanistic origin results from indirect observations, often when the cell lineages have been subject to rigorous selective pressure during development. Here, we applied haplarithmisis to infer the haplotypes and the copy number of parental genomes in 116 single blastomeres comprising entire preimplantation stage bovine embryos (n=23) following in vitro fertilization. Not only abnormal fertilizations leading to triploid zygotes, but also normally fertilized zygotes can spontaneously segregate entire parental genomes into different cell lineages during cleavage of the zygote. We coin the term “heterogoneic division” to indicate the events leading to non-canonical zygotic cytokinesis segregating the parental genomes into distinct lineages. Persistence of those cell lines during development is the likely cause of chimaerism and mixoploidy in mammals.
