Investigate changes in gene expression and enriched pathways in endothelial cells following Liver endothelial Heg1 knockout.

ABSTRACT: Knockout of Heart-of-glass (Heg) in liver endothelial cells alters the metabolic patterns of hepatocytes. To further investigate whether endothelial cells regulate hepatocyte function and gene expression patterns by secreting certain proteins or factors mediated by Heg1, we collected mice at postnatal days 3 (P3) and 42 (P42), isolated liver endothelial cells, and performed RNA-seq analysis to explore the interaction patterns between endothelial cells and hepatocytes.

Stellate cells, hepatocytes and endothelial cells imprint the Kupffer cell identity on monocytes colonizing the liver macrophage niche (microarray)

Project description:Macrophages are strongly adapted to their tissue of residence. Yet, we know little about the cell-cell interactions that imprint the tissue-specific identities of macrophages in their respective niches. Using conditional depletion of liver Kupffer cells, we traced the developmental stages of monocytes differentiating into Kupffer cells and mapped the cellular interactions imprinting the Kupffer cell identity. Kupffer cell loss induced the tumor necrosis factor (TNF) and interleukin-1 (IL-1) receptor-dependent activation of stellate cells and endothelial cells, resulting in the transient production of chemokines and adhesion molecules orchestrating monocyte engraftment. Engrafted circulating monocytes transmigrated into the perisinusoidal space, and acquired the liver-associated transcription factors ID3 and LXRα. Coordinated interactions with hepatocytes induced ID3 expression, while endothelial cells and stellate cells induced LXRα via a synergistic NOTCH-BMP pathway. This study shows that the Kupffer cell niche is composed of stellate cells, hepatocytes and endothelial cells that together imprint the liver-specific macrophage identity.

Project description:In this present study we demonstrated portal hypertension after PHx promoted liver regeneration by activating mechanoreceptor Piezo1 in endothelial cells. Next, in vivo and in vitro experiment proved that Piezo1 activation in LESCs enhanced liver regeneration via Wnt2/β-catenin mediated LESCs-hepatocyte crosstalk: Shear stress-induced Piezo1 activation in LESCs promoted secretion of Wnt2 protein to the liver microenvironment via AFT2/ID1 signaling pathway, which further promoted the nuclear translocation of β-catenin in hepatocytes and consequently enhanced the proliferation of hepatocytes (Fig. 6M). In addition, we confirmed the translational value of Piezo1 agonist Yoda1 in promoting liver regeneration after PHx.
