Trancriptomic profilin of HLMVECs following catecholamine exposure

ABSTRACT: This study presents bulk RNA sequencing data from human lung microvascular endothelial cells (HLMVECs) exposed to defined concentrations of catecholamines for 4 h and 24 h. The dataset was generated to characterize endothelial transcriptional responses to sustained adrenergic stimulation under controlled in vitro conditions.

Project description:The two-component system qseBC is a putative prokaryotic adrenergic receptor. In Aggregatibacter actinomycetemcomitans (Aa), it was shown that the qseBC responds significantly to the presence of both catecholamines and ferrous iron. In addition, growth is significantly increased in the presence of both. Therefore, we performed a custom Aa microarray in order to determine the effects of catecholamines and catecholamines and iron (CAT-Fe) on Aa. Aggregatibacter actinomycetemcomitans was grown in triplicate in a chemically defined media (CDM) supplemented with either 100uM of ferrous chloride or 50uM of norepinephrine or both. At mid-log, RNA was harvested using the Qiagen Lipid Tissue Minikit.

Transcriptomic analysis of a zebrafish model of heart failure induced by isoproterenol

Project description:Heart failure is a complex cardiac disorder characterized by ventricular dilation and disruption of the cardiac performance. After a cardiac injury, such as a myocardial infarction, the activation of sympathetic and adrenergic systems attempts to maintain cardiac output. However, sustained elevation of circulating catecholamines triggers maladaptive changes in the heart that eventually lead to heart failure. Chronic isoproterenol treatment is a well established approach to mimic the development of heart failure in animal models to study pathophysiological mechanisms.

Eukaryotic catecholamine hormones influence the chemotactic control of Vibrio campbellii by binding to the coupling protein CheW

Project description:In addition to their well-known role as stress-associated catecholamine hormones in animals and humans, epinephrine (EPI) and norepinephrine (NE) act as interkingdom signals between eukaryotic hosts and bacteria. However, the molecular basis of their effects on bacteria is not well understood. In initial phenotypic studies utilizing Vibrio campbellii as a model organism, we characterized the bipartite mode of action of catecholamines, which consists of promotion of growth under iron limitation, and enhanced colony expansion. In order to identify the molecular targets of the hormones, we designed and synthesized tailored probes for chemical proteomic studies. As the catechol group in EPI and NE acts as iron chelator and is prone to form a reactive quinone moiety, we devised a photoprobe based on the adrenergic agonist phenylephrine (PE), which solely influenced colony expansion on soft agar. Using this probe, we identified CheW, located at the core of the chemotaxis signaling network, as a major target. In vitro studies confirmed that EPI, NE, PE, as well as labetalol, a clinically applied antagonist, bind to purified CheW with affinity constants in the sub-micromolar range. In line with these findings, exposure of V. campbellii to these adrenergic agonists affects the chemotactic control of the bacterium. This study highlights a previously unknown effect of eukaryotic signaling molecules on bacterial motility.

Beta3-Adrenergic receptor downregulation leads to adipocyte catecholamine resistance in obesity

Project description:This project investigates mechanisms underlying catecholamine resistance in obesity. We found that adipocyte Beta3-adrenergic receptor (Adrb3), the dominant isoform mediating lipolysis, undergoes transcriptional downregulation after ligand exposure or inflammatory stimulation, representing homologous and heterologous desensitization, respectively. Both processes are mediated by the EPAC-RAP2A-PI-PLC-TRIB1 signaling axis, which promotes degradation of the transcription factor C/EBP-alpha and suppresses Adrb3 expression. Pharmacologic inhibition of EPAC/RAP restored Beta3-adrenergic responsiveness, enhanced lipolysis, and improved energy expenditure in obese mice. These findings reveal a molecular pathway contributing to catecholamine resistance and impaired energy mobilization in obesity.

A catecholamine-independent pathway controlling adaptive adipocyte lipolysis

Project description:Several adipose depots, including constitutive bone marrow adipose tissue, resist conventional lipolytic cues. However, under starvation, wasting or cachexia, the body eventually catabolizes stable adipocytes through unknown mechanisms. Here we developed a mouse model of brain-evoked depletion of all fat, including stable constitutive bone marrow adipose tissue, independent of food intake, to study this phenomenon. Genetic, surgical and chemical approaches demonstrated that catabolism of stable adipocytes required adipose triglyceride lipase-dependent lipolysis but was independent of local nerves, the sympathetic nervous system and catecholamines. Instead, concurrent hypoglycaemia and hypoinsulinaemia activated a potent catabolic state by suppressing lipid storage and increasing catecholamine-independent lipolysis via downregulation of cell-autonomous lipolytic inhibitors including G0s2. This was also sufficient to delipidate classical adipose depots and was recapitulated in tumour-associated cachexic mice. Overall, this defines unique adaptations of stable adipocytes to resist lipolysis in healthy states while isolating a potent catecholamine-independent neurosystemic pathway by which the body can rapidly catabolize all adipose tissues.

RNA-seq of cultured human kidney peritubular microvascular endothelial cells following exposure to cyclosporine A

Project description:This study represents the first time human kidney endothelial cells were exposed to cyclosporine A (CsA), a calcineurin inhibitor known to contribute to nephrotoxicity with symptoms including microvascular injury. This toxicity has been found to be unique to human trials and human kidneys are particularly susceptible to injury. We found that human kidney peritubular microvascular endothelilal cells (HKMECs) that were exposed to CsA exhibited transcriptomic changes around genes important to VEGF signaling and endothelial inflammation.
