Project description:Adrenarche, characterized by rising adrenal dehydroepiandrosterone (DHEA) levels, precedes puberty and has been implicated in reproductive maturation, and disorders like polycystic ovary syndrome (PCOS). However, the mechanisms by which DHEA influences the hypothalamic-pituitary-gonadal (HPG) axis remain poorly understood, largely due to the absence of an appropriate animal model. The spiny mouse (genus Acomys), which has a prolonged juvenile phase and endogenous prepubertal DHEA production, provides a valuable system to investigate the effects of early-life exposure to DHEA. We confirmed the spiny mouse prepubertal rise in circulating DHEA levels, which occurs independently of HPG axis activation. Transcriptomic analysis revealed increased ovarian expression of steroidogenic and estrogen-responsive genes, suggesting possible effects of DHEA. We show in vitro that DHEA induces Prlr and Prl expression seemingly via the estrogen receptor. Adrenalectomy (Adx) followed by DHEA “rescue” (AdxDHEA) revealed that DHEA can be synthesized and metabolized outside the adrenal glands. Ovarian transcriptomes from AdxDHEA mice exhibited extensive changes, including upregulation of immune response-related genes, and downregulation of cholesterol-metabolism and estrogen-synthesis genes, most notably Cyp19a1. The reduced Cyp19a1 expression is likely due to elevated Amhr2, and was accompanied by a high androgen-to-estrogen ratio, indicating a DHEA-induced, PCOS-like phenotype. The rise in DHEA during an extended juvenile phase establishes the spiny mouse as a suitable model for studying the consequences of elevated DHEA in childhood. Our findings reveal that prepubertal exposure to DHEA alters ovarian gene expression and steroidogenesis, providing mechanistic insight into how early androgen excess may predispose to reproductive dysfunction.

Project description:The disorders of endometrial receptivity and ovulatory dysfunction are both important causes of infertility of patients with Polycystic Ovary Syndrome (PCOS). The study aimed to investigate the expression profile and functional analysis of circRNAs in the endometrial receptivity of mice with PCOS. Twenty-four 4-week-old female C57BL/6 mice were randomly divided into two groups (PCOS group and Normal control group, n=12) and PCOS group was subcutaneously with DHEA 6mg/kg for 20 days. The circRNA expression profile in the endometrium tissue of two groups on the 4th day of gestation was screened by gene chip technique.

Project description:To identify a role for protein palmitoylation in ovarian of DHEA-induced PCOS mice model. First, we used DHEA to induce a PCOS mice model，hematoxylin-eosin (H&E) staining sections form the ovaries of the Control and DHEA groups.The mice oral gavage with DHEA disrupted ovarian morphology,the estrous cycle, and hormone profilein DHEA-induced mouse. The three pairs of ovary for each tube are mixed together as a group were lysed and divided into 2 fractions. The -HAM condition served as a negative control and the +HAM sample positive control.In the +HAM sample, the palmitate residue was cleaved off and exchanged with biotin. After the ABE reaction was completed, streptavidin beads were used to enrich for biotinylated proteins. Proteins enriched from +HAM and −HAM conditions were identified using mass spectrometry (MS).

LNCaP cells were exposed to DHEA (300 nM), DHT (300 nM), or vehicle for 48 hrs, and mRNA was isolated. mRNA expression was measured using Affymetrix HU-95 gene chips in 3 experiments performed on different dates. Gene expression values for the two treatment groups and control were sorted in ascending order on the p-values corresponding to a variance stabilized Hotelling test, which measured the extent of differential RNA expression between control and either hormone treatment. The top four genes with significant differential expression were S100 calcium binding protein, neurotensin, 24-dehydrocholesterol reductase, and anterior-gradient 2 homologue. Corresponding per comparison p-values were less than 3 x 10 -5. Nested tests of differential expression between DHEA and DHT treatment revealed significant differences (p < 0.01) for two of the four genes: the S100 calcium binding protein and neurotensin. The microarray findings were confirmed by quantitative RT-PCR. The top 83 genes found to exhibit differential expression were used in a pathway analysis. In general, DHT decreased expression of more genes involved in intercellular communication, signal transduction, nucleic acid binding and transport, and in structural components, such as myosin and golgin, than did DHEA. These data reveal consistent, measurable differences in gene expression patterns following treatment of LNCaP prostate cancer cells with DHEA versus DHT. The mechanisms underlying these observations, and the possible pathophysiological significance of these differences, remain to be determined. Experiment Overall Design: Three separate experiments were performed and labeled: 111502, 011403, and 022803. Three groups were run per experiment: Control, DHT or DHEA. Cells were exposed for 48 hr to 300 nM of DHT or DHEA. One Affymetrix HG_U95Av2 microarray chip per group was analyzed. The data was normalized by the "dchip" process.

Project description:Aging is associated with systemic chronic inflammation (inflammaging) that leads to impaired physiological functions and vulnerability to several diseases. However, underlying alterations in aged immune system resulting in gradual loss of immune fitness remain unclear. Using a combination of CD8 T cell transfer from young to old and from old to young mice and single-cell RNA sequencing, we characterized the age-associated alterations in CD8 T cells. We transferred 2 millions of purified CD8+ T cells pooled from 3 CD45.1 C57BL/6J 3 months old female mice (donor) by i.v. injection into CD45.2 C57BL/6J 24 months old female mouse (recipient) and sorted CD45.1+CD3e+CD8a+ and CD45.2+CD3e+CD8a+ T cells from the spleen 1 month post transfer to perform scRNA/TCR-seq.
